This paper discusses in detail the beeswax as raw materials preparation andpurification of twenty-eight alkanol process, and then carry out the process of theoptimization; At the second, Octacosanol for the physiologic function of SD rats andSD rats blood biochemical parameters, hormone secretion followed by SD rats as theexperimental model ; Finally, by the molecular biology experiments to researchOctacosanol involved in energy metabolism gene expression in SD rats body ,in-depth study of the mechanism for the regulation of energy metabolism in rats .In experiment 1: At the first of the Experimental , Refined beeswax alcoholrelative to saponification treatment, saponification during the preparation of therequired saponification temperature, saponification time, the concentration of NaOHethanol solution , the solid-liquid ratio four single factor level for the process ofsaponification extract , identified in a single factors under the best conditions; Byorthogonal experimental design to determine the optimal saponification conditions ,temperature 95°C for saponification , saponification time 3.5h, NaOH concentrationof ethanol solution of 8% , the solid-liquid ratio of 1:8 , the saponification conditionsoptimal . Then purified through molecular distillation technology can be obtained ofthe highest purity of 89.78% .In experiment 2: Using SD rats as a model added different concentrations ofOctacosanol in the feed, study the effects of the SD rats on growth performance,physiological and biochemical parameters , hormone secretion.The experiment used48 healthy SD rats ( male and female ) , in accordance with the principles of weightconsistent divided into 4 groups : experimental group 1 (saline controlgroup) ,experimental group 2 ( high Octacosanol concentration group) , experimentgroup 3 (medium Octacosanol concentration group), experimental group 4 ( lowOctacosanol concentration group ).The rat can feed and drink distilled water byitsleves, the test time is 30 days.Record the intake of the feed , weight and otherindicators .At the end of the experiment collect of the rat blood,liver,muscle and other tissue samples.Then test the fluorescence quantitative GLUT-4,STAR,GS,STAR andmany other genes expression. Test results showed:1.Relative to the saline blank control group fed with high , medium and lowthree different concentrations of octacosanol emulsifier significantly improve the SDrats of body weight , daily weight gain , physiological and biochemical parameterssignificantly ; the SD The rat growth hormone , glucagon secretion were significantlyincreased.2.Relative to saline blank control group fed the three different concentrations ofoctacosanol,The emulsifier, total cholesterol, LDL significantly reduced high-densitylipoprotein increased significantly ;And the third experimental group had a best work.3.Relative to saline blank control group fed with high , low concentration ofOctacosanol.The Content of GLUT-4 , STAR, GS AMPK gene expression levels in ratliver and muscle tissue were higher than control group , and the third experimentalgroup expression was increased in the most obvious.
