Effects Of Cellular Immunity,IL-2and TNF-α By Opioid Drugs On Hepatic Cancer Mice

Objective:To study the effects of cellular immunity, IL-2and TNF-α caused by Opioid drugs on hepatic cancer mice.Methods:The models of H22hepatic cancer mice were established by the cell inoculation method, and were randomly divided into5groups:group Saline、group Morphine、group Pethidine、group Fentanyl and group Sufentanil。10rats in each group were picked up randomly and injected Opioids once a day for3consecutive days when7days after built models of H22hepatic cancer mice。Group Saline was followed intraperitoneal injection of0.5ml Saline, group Morphine was injected with Morphine by5mg/kg, group Pethidine was injected with Pethidine by40mg/kg, group Fentanyl was injected with Fentanyl by50μg/kg and group Sufentanil was injected with Sufentanil by5μg/kg. After24h of the last intraperitoneal injection, all mice were subjected to laparotomy and collected mice eyes blood to detect of lymphocyte subsets CD3+、CD4+CD8+and CD4+/CD8+counts, the number of IL-2and TNF-a were detected by Enzyme-linked Immunosorbent Assay (ELISA).Results:Compare to group Saline, the numbers of CD3+、CD4+、CD8+and the ratios of CD4+/CD8+showed no significant different in group Pethidine (P >0.05), but decreased significantly in group Morphine, group Fentanyl and group Sufentanil (P<0.05). Compare to group Saline, the numbers of IL-2increased significantly in group Pethidine (P<0.05), and the numbers of TNF-a showed no significant difference (P>0.05), but decreased significantly in group Morphine, group Fentanyl and group Sufentanil (P<0.05)Conclusion Morphine, Fentanyl and Sufentanil can suppress the immune function, decrease significantly IL-2and TNF-α of hepatic cancer mice。 Pethidine suppress the immune function unobviously, and the numbers of TNF-α has no significant difference, but increase significantly the numbers of IL-2.The treatment of hepatic cancer has reference value for using Pethidine.
