Objective:We investigated that the expression of moncyte chemoattrant protein-1(MCP-1) mRNA and protein by HK-2cells exposed to Calcium oxalate monohydrate and oxalate, and protective mechanism of taurine on cells.Methods:HK-2cells, a proximal tubular epithelial cell line, were cultured. Eight groups each were divided in by this study:control group; Calcium oxalate monohydrate and oxalate group; Taurine and Calcium oxalate monohydrate and oxalate group; Apocynin and Calcium oxalate monohydrate and oxalate group; Catalase and Calcium oxalate monohydrate and oxalate group; Taurine group; Apocynin group; Catalase group. After6h, the culture media was tested for LDH, H2O2,8-isoprostane, and MCP-1protein. Gene expression of MCP-1mRNA and P47phox mRNA was determined by reverse transcriptase-polymerase chain reaction.After24h, we investigated cells livability.Results:Exposure to Ox and CaOx crystals resulted in significantly decrease in cells livability, and by taurine, apocynin, catalase led to obviously increase. The LDH, H2O2,8-isoprostane, expression of MCP-1mRNA and protein were increased following exposure to Calcium oxalate monohydrate and Oxalate, which were decreased during co-treatment with taurine, or apocynin, or catalase. Exposure to Ox and CaOx crystals resulted in significantly increase in P47phox mRNA, which was not reduced with treatment of Taurine, catalase except apocynin.Conclusion:The calcium oxalate monohydrate and oxalate can stimulate the renal epithelial cells to produce oxidative stress (OS). It may lead to increase expression of MCP-1mRNA.With treatment of antioxidant and taurine, oxidative stress was alleviated and expression of MCP-1was decreased. Effect of taurine may be not in relation to reduction of P47phox mRNA.
