Part Ⅰ Effect of erythropoietin after spinal cord injury in ratsObjiectiveTo investigate the Apoptotic protein expression of P53in the exploring of erythropoietin on spinal cord injury.Methods44adult male SD rats were randomly divided into sham group A (n=4), control group B (n=20), the experimental group C (n=20), A group only Iaminectomy was made without impact trauma, B、Cgroup establish spinal cord injury(SCI) model by modified Allen’s model, EPO therapy group C peritoneal injection3h after injury(1000IU/kg), A、B group the same dose of normal saline were injected。Each group after6h、1d、3d、7d four time points through the the Tarlov score on behavioral evaluation of spinal cord function, and the expression of P53cell were detected by immunohistochemistry in spinal cord injury tissue, comparative analysis of the difference between the two groups.ResultsEPO prophylaxis group、EPO treatment group significantly much higher than the control injury group at each time point in Tarlov score (P<0.05);the expression of P53appeared both treatment、control group at6h, which peaked at3d and decreased at7d, the expression of P53cell apoptosis in experimental group was significantly less than the control group at each time point (p<0.05), the Tarlov score and the expression of P53cell apoptosis with the correlative coefficiency r=-0.5290, the difference was significant (P=0.016<0.05).ConclusionThe expression of P53was induced after spinal cord injury;Tarlov neurofunction score lower is correlated with the expression elevate of P53; erythropoietin can reduce the expression of P53, play a protection after spinal cord injury. To explore the Protective effect on neuromotor function of different time use erythropoietin in rat after spinal cord injury, as to to provide more theoretical guide for the use of erythropoietin for preservation of spinal function.MethodA total of64adult male Sprague-Dawley rats were randomly divided into four groups, sham group A(n=4)> control group B(n=20)、EPO prophylaxis group C(n=20). EPO therapy group D(n=20), A group only laminectomy was made without impact trauma, B、C、Dgroup establish spinal cord injury(SCI) model by modified Allen’s model, EPO prophylaxis group C peritoneal injection3h before injury(1000IU/kg), EPO therapy group D peritoneal injection3h after injury(1000IU/kg), A、B group the same dose of normal saline were injected。 Each group after6h、1d、3d、7d four time points through the the Basso,Beattie,and Bresnahan(BBB) scoring system and Rivlin’s tiltboard experiment on behavioral evaluation of spinal cord function, hematoxylin and eosin staining wasperformed for histological observation after injury.ResultMotor function score:EPO prophylaxis group C, EPO treatment group D significantly much higher than the control injury group B at each time point (P <0.05); at the6h time point,there was no significant difference between C、D group(P>0.05), at the1d、3d、7d time point, there was significant difference between C、D group(P<0.05)。 pathology:in group A,the noundary between gray-while matter was clear and neurons were evidenced abundant; in group B,ultiple hemorrhage and necrosis was seen in the injured spinal cord the number of neurons decreased and gradually aggravated; in group D was minor than that in group B,and the structures inside were gradually relieved; in group C was the mildest compared with group B and D,few hemorrhage was seen,the organelles in neurons swelled mildly,and the swellling almost disappeared in latter.ConculsionEPO can promote the neurofunction recovery and play a protection after spina cord injury, EPO prophylaxis group and treatment group consistent effect at6h time point, better than the treatment group at ld、3d、7d time point, It will be better than to use EPO3hour before surgery than after surgery.
