Background and purposeWhen people opened twenty-first Century gate, one of the world’s aging society also show in front of. China has gradually entered the aging society. The latest survey showed that over60years old has a population of more than127million; over the age of70with a population of more than200million.. Our country people over age60, vascular dementia accounted for3.2%, the total number of patients in3000000above. Cognitive dysfunction is cerebral degenerative lesions of the behavior, the most severe pathological state of dementia, it not only for patients with long-term pain, severely affect the quality of life. But for families and society bring heavy burden. Due to the current domestic and foreign to the control of the disease progression is the effective method and medicine, so the world of geriatric research institutions have listed it for the next10years research, our country also will be included in the national key project.Vascular dementia (vascular dementia, VaD) of the detailed pathogenesis is not completely clear, think generally, VaD and excitotoxicity, inflammation, free radicals, cholinesterase and apoptosis, mitochondrial dysfunction and other theories about. In recent years the study found that endogenous nitric oxide, carbon monoxide as a neurotransmitter play a role in hippocampus in learning and memory processes in long-term potentiation.Role of heme oxygenase-1(heme oxygenase-1, HO-1) and carbon monoxide (carbon monoxide, CO) system in many physiological and pathological processes play an important role in the organization, especially for the organ protective effects. In ischemic brain injury, HO-1/CO system has antioxidant, stabilization of cell membrane and mitochondria membrane, inhibiting cell apoptosis. HO-1/CO system protection effect is in the affirmative, but in the VaD rat model, whether exercise training on HO-1/CO system influence and the rehabilitation training to improve VaD rat cognitive function of the specific mechanism is not clear, whether through the effects of rehabilitation training on HO-1expression and to improve the function of learning and memory, it is this study reports, through the establishment of vascular dementia rat model, to observe the expression change of HO-1, discuss rehabilitation training in the treatment of VD specific mechanismsMethodsResearch is divided into two parts. The first part, using SD rats as the research object, adopts a permanent bilateral common carotid artery ligation followed from the middle shearing method to prepare the VaD animal model, the experimental animal were randomly divided into the rehabilitation training group, control group, sham operation group; modeling success after rehabilitation training group for rehabilitation training, lasting4weeks, weekly6D training, training1times a day. The control group and sham operation group were not rehabilitation training. Rehabilitation training after application of MG-2Y-type maze test in rats learning and memory ability test. Experiment in a quiet dark environments. Rats by electric shock after one-time starting from the area fled to safety in the reaction zone to" correct responses", or" error response". Learning ability:rats subjected to electric shocks from the district started to jump directly to the safety zone for correct responses, to reach10consecutive trials9times can make the correct responses before the required number of electric shocks, i.e. the error response times (error number, EN) as part of their academic achievement. The number of electric shocks that required less learning ability. Memory capacity:the standard rats at rest after24h, the same method for the detection of memory, using a rat10shock in the correct response times (correct number, CN) as a result of the memory. The second part using HE staining, HO-1mRNA levels were measured, and compare the difference.Results1, Y-type maze test:compared with sham operation group, rehabilitation training group and the control group learning and memory performance in both inferior and sham operation group. Rehabilitation training group compared with the control group, results suggest that rehabilitation training on learning and memory ability can be improved.2, pathology detection:in hippocampus CA1region HE staining were visible: rehabilitation training group:hippocampal neuronal number, level is clear, structure is arranged in order, the nucleus, nucleolus chromatin and abundant, clear, a small nuclear pyknosis degeneration of neurons. Control group:the reduction in the number of neurons in the hippocampus, contour, arranged in loose structure, cell gap widened, the nucleus has a smaller volume, dyeing depth, a nuclear pyknosis performance. Sham operation group:hippocampus neuron number is rich, the cell contour clear, tidy boundary, structure, the nucleolus is clear, uniform dyeing.3, the expression of HO-1mRNA results:Semi-quantitative RT-PCR results show, groups are visible on HO-1mRNA expression, rehabilitation training group and the control group HO-1mRNA expression than the sham operation group, rehabilitation training group was higher than that of the control group. Electrophoretic band analysis shows, rehabilitation training group the expression of HO-1mRNA transcription level is the highest, the control group, sham operation group minimum.Conclusion1, VaD HO-1mRNA expression in rat hippocampus of rats decreased, and the ability of learning and memory impairment in relation to.2, rehabilitation training is possible through the increased expression of HO-1mRNA, improve the ability of learning and memory in VaD rats.
