Objective Respectively by the groups of vascular dementia rats(vascular dementia,VD)for electric acupuncture and rehabilitation and rich acusector joint treatment of rehabilitation training,observe the comparison between groups of VD rats memory,ability to learn,as well as to the integrin alpha v beta 3 and vascular endothelial growth factor(vascular endothelial growth factor,VEGF)expression.From the perspective of angiogenesis,this paper discusses the curative effect of the above treatment on vascular dementia,and provides important data for the treatment of vascular dementia with acupuncture and rehabilitation.MethodsUsing two vascular blocking of VD model rats,choose clean level male Sprague wrote-Dawley(SD)rats,90,through the water maze experiment on the screen,select two vessels permanent ligation method(2-vessle occlusion,2-VO)copy the vascular dementia rats model,and combined water maze and preparation of neurobehavioral evaluation model is successful,the rats with dementia were divided into normal group,model group,sham operation group,electroacupuncture group,rehabilitation group and acupuncture group.Normal group,model group,sham operation group did not give any treatment,electric acupuncture group was treated with electric acupuncture(will,the da-zhui acupuncture point),group with abundant rehabilitation environment(with more rat cage,cage put coloured toys)and rehabilitation training(stick,screen,the balance beam),needle kang group by electric acupuncture combined rehabilitation therapy,treatment once a day,the rest two days a week,between groups in treatment of 28 days again in water maze test and put to death.The water maze was used to test the learning ability and memory improvement of VD rats;the TTC staining was used to detect the area of cerebral ischemia infarction in rats;The HE staining was used to detect the changes of cerebral cortex and hippocampal pathology in rats;TheImmunohistochemistry was used to detect the expression of VEGF in rat hippocampal and cortical VEGF conditions;The Enzyme linked immunosorbent assay(ELISA)was used to detect the changes of VEGF and alpha CD34 immunohistochemistry was used to detect the angiogenesis of capillaries.Results1.There was no significant difference between the rat model group,the electroacupuncture group,the rich rehabilitation training group and the acupuncture group.After treatment,the average escape latency of the rats was significantly shortened in the electroacupuncture group,the rich rehabilitation training group and the acupuncture group,and the comparison was statistically significant(P<0.01).After treatment,the number of electroacupuncture group,rich rehabilitation training group and needle kang group were more than that of the model group,with statistical difference(P<0.01).The average latency period of the rats in each treatment group was higher than that of the acupuncture group and the rich rehabilitation training group was longer than that of the acupuncture group,and the comparison was statistically significant(P<0.05).After treatment,the number of acupuncture groups and rich rehabilitation training groups of 28 days after treatment was less than that of the acupuncture group,and the comparison was statistically significant(P<0.05).The average avoidance latency and the number of crossing platforms in the 28 days after treatment were not statistically significant(P> 0.05)compared with the electroacupuncture group and the rich rehabilitation training group.2.Cerebral infarction volume detection: normal group and control group without infarcts,infarcts model group was obviously,after intervention treatment,the treatment group of cerebral infarction volume smaller,compared with model group,electric acupuncture group,rich rehabilitation training and needles,cerebral infarction volume significantly narrowed,statistically significant difference(P<0.05 or P<0.01);Compared with the electroacupuncture group and the rich rehabilitation training group,the volume of cerebral infarction in the acupuncture group was significantly smaller than that of the electroacupuncture group and the rich rehabilitation training group,and the difference was statistically significant(P<0.05).There was no significant difference between the electroacupuncture group and the rich rehabilitation training group(P>0.05).3.Histopathological observation: Groups of rats were normal group and the control group,the brain HE dyeing uniformity,number of hippocampus CA1 area neurons in normal,organization,distinct,nerve cells in the normal structure shape,the completeness of the envelope and nucleolus is clear,no nerve interstitial edema;In the rat brain of the model group,the edema of the bilateral hippocampal area was obvious,the boundary of the cell membrane and the nuclear membrane was unclear,the nucleus was deeply stained,and the gap of the cells was widened,arranged disordered,and the nucleoli disappeared.Three groups with different levels of treatment group rats had nerve edema,neuron populations,cell shape to improve the situation,the needle kang group than other treatment group of rats had the potential to improve the more apparent,neat cells and abundant cytoplasm and deformation,necrosis significantly reduced the number of neurons.4.Expression of positive cells of VEGF and integrin alpha 3: in the normal group and the sham operation group,there was almost no expression of the integrin in the hippocampal CA1 region,and the VEGF was scattered in expression.In the model group,VEGF and integrin in the hippocampal CA1 area were increased,and the average optical density was significantly increased,and there was a significant difference between the normal group and the sham group(P<0.01 or P<0.05).Each treatment group by electric acupuncture and rehabilitation training rich and abundant acusector joint rehabilitation training after intervention therapy,the rat hippocampal CA1 area of VEGF,integrin alpha v beta 3 average optical density value compared with the model group were significantly higher and the difference is statistically significant(P<0.01 or P<0.05);There was no significant difference between the average optical density and the rich rehabilitation training group(P>0.05).The average optical density of VEGF and integrin in the acupuncture group was significantly higher than that of the electroacupuncture group and the rich rehabilitation training group(P<0.01P<0.05).5.New blood vessel detection: the CD34 immunohistochemical method was used to mark the microvascular endothelial cells.In the light microscope,the newborn microscopic blood vessels were stained brown,forming a brown tube cavity.There were fewer microvascular expression in rats in normal group and sham operation group.Compared with the normal group and the sham operation group,the microvascular increase in the model group was statistically significant(P<0.01).The microvascular expression of the electroacupuncture group,the rich rehabilitation training group and the needle kang group was further increased on the basis of the model group,and the comparison with the model group was statistically significant(P<0.05 or P<0.01).Compared with the electroacupuncture group and the rich rehabilitation training group,the number of microvessels increased more and the difference was statistically significant(P<0.05 or P<0.01).6.VEGF and integrin protein expression in alpha beta 3 v: normal group and control group of VEGF,integrin alpha beta 3 v content is low,the model group rats VEGF,integrin alpha v beta 3 content increased significantly compared with significant difference(P<0.01);Each treatment group by electric acupuncture and rehabilitation training,as well as rich rich acusector joint rehabilitation training after intervention therapy,VEGF in rats blood,integrin alpha v beta 3 levels compared with model group were significantly higher and the difference has statistics meaning(P<0.01 or P<0.05);There was no significant difference between the electroacupuncture group and the rich rehabilitation training group,VEGF,and the integrin alpha and alpha 3(P>0.05).The content of VEGF and integrin in the needle kang group was significantly higher thanthat of the electroacupuncture group and the rich rehabilitation training group(P<0.01 or P<0.05).Conclusion1.Rich acusector joint rehabilitation therapy can promote ischemic vascular dementia rats angiogenesis promotes the repair of damaged neurons,improve VD rats learning and memory function,and the effect is better than that of pure electric acupuncture treatment and rehabilitation training.2.The combination of electroacupuncture and rich rehabilitation training can improve the expression of VEGF and integrin aV beta 3,and the effect is better than single electric acupuncture and rich rehabilitation training.3.Electroacupuncture combined with rich rehabilitation therapy can reduce the volume of cerebral ischemia infarction in rats with vascular dementia,which is better than simple electroacupuncture therapy and rich rehabilitation training.4.Rich acusector joint rehabilitation treatment to improve the mechanism of vascular dementia rats learning and memory function may be to promote angiogenesis factor by raising(VEGF and integrin aV beta 3)expression,by increasing the cerebral blood flow to promote restoration of the neurons.5.Rich acusector joint rehabilitation therapy compared with single use electric acupuncture or rich rehabilitation training may in the improvement of cognitive function of VD rats has more advantages,suggested that clinical should further promote the comprehensive therapy.
