Background and objectivesGastric cancer is a common malignant tumor in the digestive system.It is the fourth most common cancer, and its mortality is the second of various malignant tumor around the world.The morbidity and mortality of gastric cancer is the first of various malignant in Asian countries,such as China, Japan, South Korea and other developing countries.It is one of the disease that is a serious threat to hunman health and life.The symptoms of gastric cancer and pre-cancerous are hidden and nonspecific,so early gastric cancer is difficult to find. When the clinical symptoms is obvious,the lesion is already late.The overall 5-year survival rate is less than 30%.In recent years,comprehensive treatment of gastric cancer has achieved a great improvement, but the treatment prospects of patients is still not optimistic.Early diagnosis and anticipation the risk of metastasis are very critical to improve the prognosis of gastric cancer patients.Therefore, it is very significance for us to find a meaningful pathology indieators which can indicate the evolution and prognosis of gastric cancer.FBXW7 is one of F-box protein family members. It can identify the target proteins of the SCF ubiquitin ligase. Recent studies show that FBXW7 is a tumor suppressor factor.The deletions and mutations of FBXW7 have been found in a variety of tumor cells. SRC-3 is one of the p160 steroid receptor coactivator family members.As a new proto-oncogene,the over-expression of the SRC-3 protein plays an important role in the development process of various tumors.KLF5 is a transcription factor of eukaryotic zinc finger protein in recently yeas.It is one of the Spl-and Kruppel-like transcription factors family members.It plays an important role in many physiological and pathological processes,including cell growth,differentiation,cell cycle regulation, angiogenesis and inflammatory response. In this study,we investigate the expression of FBXW7,SRC-3,KLF5 in gastric cancer and their relationship with clinicopathological parameters in the development process of. gastric cancer.These provide new ideas to fing valuable clinicopathological indicators and new targets for cancer therapy.Materials and Methods160 case of gastric carcinoma tissue,25 cases of para-carcinoma atypical hyperplasia tissues and 20 cases of para-carcinoma normal mucosa tissue were collected.All patients were not receiving preoperative radiotherapy, chemotherapy or biological targeted therapy. In the 60 cases of gastric carcinoma,50 cases of adenocarcinoma,10 cases of signet ring cell carcinoma; 12 cases of well-differentiated carcinoma,24 cases of moderately differentiated carcinoma,24 cases of poorly differentiated carcinoma; 29 cases with no lymph node metastasis,31 cases with lymph node metastasis;TNM staging:28 cases of stageâ… and stageâ…¡,32 cases of stage III and stage IV.2 The expression of FBXW7,SRC-3 and KLF5 proteins were detect by SP immunohistoehemistry.The clinicopathologic parameters of gastric carcinoma and the correlation among the three indicators were analyzed,and the significance of the three indicators were explored in gastric carcinoma.3 Statistical analysis:All the data were analyzed by SPSS 17.0 software. Chi-square test was used to analyze the date.The correlation of FBXW7,SRC-3 and KLF5 was analyzed with Spearman rank correlation.Test level a=0.05.results1 The positive rates of FBXW7 in gastric carcinoma, atypical hyperplasia tissues and normal tissue were 46.7%,64.0%,90.0%,the difference among the three groups was statistically significant(P<0.05). The expression of FBXW7 in gastric carcinoma and atypical hyperplasia tissues was significantly higher than the normal tissue(P<0.05),but there was no significant different between the former two groups(P>0.05).The expression of FBXW7 was significantly correlated with differentiation degree, lymph node metastasis and clinical stage(JP<0.05).2 The positive rates of SRC-3 in gastric carcinoma, atypical hyperplasia tissues and normal tissue were 48.3%,32.0%,10.0%,the difference among the three groups was statistically significant(P<0.05). The expression of SRC-3 in gastric carcinoma was significantly higher than the normal tissue(P<0.05),but there was no significant different between atypical hyperplasia tissues and gastric carcinoma,normal tissue(P>0.05).The expression of SRC-3 was significantly correlated with depth of invasion, lymph node metastasis and clinical stage(P<0.05).3 The positive rates of KLF5 in gastric carcinoma, atypical hyperplasia tissues and normal tissue were 66.7%,44.0%,15.0%,the difference among the three groups was statistically significant(P<0.05), and statistical difference was also get from the two inter-group comparisons(P<0.05).The expression of KLF5 was significantly correlated with differentiation degree, lymph node metastasis and clinical stage(P<0.05).4 The correlation of the expression of FBXW7,SRC-3 and KLF5 in gastric carcinoma was analyzed statistically.There is a negative correlation between FBXW7 and SRC-3,KLF5(P<0.05).There is a positive correlation between SRC-3 and KLF5(P<0.05).Conclusions1 The expression of FBXW7 in normal tissue and atypical hyperplasia tissues was significantly higher than gastric carcinoma.The expression of FBXW7 was significantly correlated with differentiation degree, lymph node metastasis and clinical stage.The down-regulated of FBXW7 protein may participate in occurrence and development of gastric carcinoma,and be closely related to the degree of malignancy and metastasis of gastric cancer.2 The expression of SRC-3 in gastric carcinoma was significantly higher than the normal tissue.The expression of SRC-3 was significantly correlated with depth of invasion, lymph node metastasis and clinical stage.SRC-3 may participate in occurrence and development of gastric carcinoma,,and also promote the invasion and metastasis of gastric carcinoma.3 The expression of KLF5 in normal tissue,atypical hyperplasia tissues and gastric carcinoma present a upgraded trend. The expression of KLF5 was significantly correlated with differentiation degree, lymph node metastasis and clinical stage. The high expression of KLF5 protein may be an early event in the process of gastric carcinogenesis, and be closely related to the degree of malignancy and metastasis of gastric cancer.4 There is a significant correlation between FBXW7, SRC-3 and KLF5 in gastric carcinoma.The abnormal expression of FBXW7,SRC-3 and KLF5 might play a important role in the evolution of of gastric carcinoma.
