| Objective:To evaluate LPA2 and KLF5 protein expression pattern in normal gastric tissue,gastric intraepithelial neoplasia(GIN)tissue and gastric adenocarcinoma tissue.Analyzed the association between the protein expression pattern and clinical pathological features in gastric adenocarcinoma patients.Analyzed the correlation between LPA2 and KLF5 protein expression in gastric adenocarcinoma.And further assess the effect of LPA2 and KLF5 on carcinogenesis,development and patients’ prognosis.Methods:Fifty-one gastric adenocarcinoma tissue,twenty-one GIN and thirteen normal gastric tissue were collected to test LPA2 and KLF5 expression by tissue microarray and immunohistochemistry assay.LPA2 and KLF5 positive expression rate between gastric adenocarcinoma tissue,GIN and normal gastric tissue were compared.And we analyzed the relationship between their expression and prognosis in gastric adenocarcinoma.Results:1.LPA2 mainly expressed in the cytoplasm and cell membrane,brown granular.The positive expression rate was 89.5%(17/19),81.3%(13/16),25.0%(4/16),33.3%(3/9),25.0%(3/12)and 23.1%(3/13)for poor differentiation gastric adenocarcinoma,moderate differentiation gastric adenocarcinoma,well differentiation gastric adenocarcinoma,high grade GIN,low grade GIN and normal gastric tissue respectively.The positive rate in cancer tissue was significant higher than that of GIN and normal gastric tissue with statistical difference(χ2 =29.9,P<0.001).2.LPA2 positive expression rate was 33.3%(3/9)and 25.0%(4/16)in high grade GIN and well differentiation gastric cancer with no statistic difference(P>0.05).3.LPA2 positive expression had no correlation with the age,gender,tumor location,tumor diameter and tissue grading(P>0.05).However,LPA2 positive expression was associated with the tumor invasion depth,Lauren type,vascular invasion,local lymph node metastasis and clinical stage(P<0.05);The positive rate in patients with infiltrating serous layer,Lauren diffuse type,positive vascular invasion,lymph node metastasis and advanced stage was statistical higher than that of negative patients.4.KLF5 mainly expressed in the cytoplasm,brown granular.The positive expression rate was 78.9%(15/19)、75.0%(12/16),75.0%(12/16),66.7%(6/9),58.3%(7/12)and 38.5%(5/13)for poor differentiation gastric adenocarcinoma,moderate differentiation gastric adenocarcinoma,well differentiation gastric adenocarcinoma,high grade GIN,low grade GIN and normal gastric tissue respectively.The positive rate in cancer tissue,GIN and normal gastric tissue was not statistical different(χ2 =7.35,P>0.05).5.KLF5 positive expression had no correlation with the age,gender,tumor location,tumor diameter,tissue grading,invasion depth,Lauren type,vascular invasion,lymph node metastasis and clinical stages(P>0.05).6.The median survival time was 11.02 months and 23.04 months for LPA2 positive and negative patients with statistical difference(HR=1.89,95%CI:1.11-3.63,P<0.05).7.The median survival time was 10.7 months and 12.1 months for KLF5 positive and negative patients with no statistical difference(HR=1.11,95%CI:0.53-2.363,P>0.05).8.There was positive correlation between KLF5 and LPA2 expression rate in gastric adenocarcinoma(r= 0.66,P<0.05).Conclusion:1.High expression rate is found in gastric adenocarcinoma tissue,and LPA2 positive expression is associated with the tumor invasion depth,Lauren type,vascular invasion,local lymph node metastasis and clinical stage.It suggests that LPA2 may be involved in the occurrence of gastric adenocarcinoma.Furthermore,LPA2 may be an important marker for the development and metastasis of gastric adenocarcinoma.2.The survival time that LPA2 positive patients in gastric adenocarcinoma is lower than negative patients,which indicated LPA2 may become a useful target for prognosis in gastric adenocarcinoma.3.The positive expression of KLF5 and LPA2 in gastric adenocarcinoma is positive correlation.It suggests that they may have an interrelated mechanism in gastric adenocarcinoma development. |
