Effect Of Lipopolysaccharide Preconditioning On Hepatic Insulin Resistance By Fructose Diet

Objective:To investigate the effect of Lipopolysaccharide preconditioning on the hepatic insulin resistance in mices with high-fructose diet.Methods:Part I Model. The mice were randomly divided into control group, the fructose group, endotoxin tolerance group; Control group, feeding a normal diet and tap water, Fructose group and the endotoxin tolerance group are the same diet with the control group, but drinking water to20%fructose water, endotoxin tolerance group accept three days of continuous intraperitoneal injection of LPS2mg/kg before drinking in high-fructose, then subcutaneous LPS0.1mg/kg every other day.Weighed once a week. After12weeks the animals were sacrificed and observed the liver gross-morphological and histological. Then test the fasting plasma glucose (FPG), fasting serum insulin (FINS) and insulin resistance index (FIRI); Part II:Western blotting detect suppressor of cytokine signaling (SOCS3), insulin receptor (p-IRβ/IRβ), phosphatidylinositol3-kinase P85(P-PI3KP85/PI3KP85), IL-1receptor associated kinase,(IRAK-M),immunoglobulin heavy chain binding protein (BIP), mierotubule assoeiated proteinl lighte hain3(LC3) protein expression in the liver tissue.Results:The FBS, FIRI, liver index of fructose group were significantly h-igher than normal group(P<0.05).FINS compared with the normal group also increased, no significant. Endotoxin group compared with the fructose group, FPG and FINS, FIRI, liver index were higher(P<0.05).And p-IRβ/IRβ, LC3exp-ression decreased,Other proteins SOCS3,IRAK-M and BIP increased expressi-on(P<0.05). P-PI3KP85/PI3KP85increased expression but no significant.Hep-atic steatosis was significantly worse.Conclusion:Endotoxin tolerance which is caused by chronic small doses of endotoxin subcutaneous injection increased IR.
