ObjectiveThe present study was undertaken to investigate the effects of atorvastatin and amlodipine on the expression of lectin-like oxidized low density lipoprotein receptor-1(LOX-1) and endothelial nitric oxide synthase(eNOS) in human umbilical vein endothelial cells(HUVECs) injuryed by ox-LDLMethods1. HUVECs were cultured in vitro. The cells were cultured in DMEM culture medium. Cells of passages3-5are used in experiment.2. When cells were grown to confluence, they were randomly divided into5groups:①control group:HUVECs were incubated in DMEM without any stimulators or inhibitors;②ox-LDL group:HUVECs were coincubated with50mg/L ox-LDL for24h;③atorvastatin group:HUVECs were pretreated with10umol/L atorvastatin for2h, then was incubated with50mg/L ox-LDL for24h;④amlodipine group:HUVECs were pretreated with10umol/L amlodipine for2h, then was incubated with50mg/L ox-LDL for24h;⑤atorvastatin+amlodipine group:HUVECs were pretreated with10umol/L atorvastatin and10umol/L amlodipine for2h, then was incubated with50mg/L ox-LDL for24h.3. The expression of LOX-1mRNA and eNOSmRNA were determined by real-time polymerase chain reaction (real-time PCR).Results1. LOX-1mRNA of the ox-LDL group、the atorvastatin group、the amlodipine group and the atorvastatin+amlodipine group was (3.98±0.21) times (P<0.01).(2.21±0.24) times (P<0.01).(2.46±0.14) times (P<0.01).(1.56±0.15) times (P>0.05) respectively vs the control group. Compared with the the ox-LDL group, the expression of LOX-1mRNA was decreased significantly(P<0.01). The expression of LOX-1mRNA was no difference significantly between the atorvastatin group and the amlodipine group(P>0.05).2. eNOS mRNA of the ox-LDL group, the atorvastatin group, the amlodipine group and the atorvastatin+amlodipine group was (0.346+0.021) times (P<0.01).(0.534±0.039) times (P<0.05).(0.468±0.026) times (P<0.05).(0.946±0.027) times (P>0.05) respectively vs the control group. Compared with the the ox-LDL group, the expression of eNOS mRNA was increased significantly (P<0.01). The expression of eNOS mRNA was no difference significantly between the atorvastatin group and the amlodipine group(P>0.05).Conclusion1. Ox-LDL could increase the expression of LOX-1mRNA and decrease the expression of eNOSmRNA, ox-LDL could injury the endothelial cell through this mechanism.2. Atorvastatin and amlodipine could downregulate the expression of LOX-1and upregulate the expression of eNOS injuryed by ox-LDL. Atorvastatin and amlodipine have a synergistic effect on the protective effects of blood vessels. |
