Objective:Observation alone, estrogen (a bone resorption inhibitor) or simvastatin (bone formation stimulating agent), and the sequential application of estrogen and simvastatin drugs on bone healing in bone of castrated rats with osteoporosis implant measurement Influence.Methods:Selection of32weeks old female SD rats were40, and were randomly divided into normal control group (group A) and ovariectomized group. Resection of bilateral ovaries castrated group, normal control group were only simple incision of lower abdomen skin.12weeks after ovariectomy in rat proximal tibial metaphysis implanted pure titanium screw implants and ovariectomized group were randomly divided into4groups:estrogen group (group B), simvastatin group (group C), sequential group (group D) and control group (group E) castration. Estrogen group:subcutaneous injection of estradiol benzoate and0.1mg/kg,3D dosing1times; simvastatin group:5mg/kg,1D gavage1times; sequential group:subcutaneous injection of estradiol benzoate and0.1mg/kg,3D dosing1times,2weeks later, given intragastrically simvastatin (5mg/kg d) for2weeks,5weeks of discontinuation, and application of simvastatin5mg/(kg d) by gavage for3weeks. Castrated controls:simple feeding, no drug intervention. Implantation after12weeks all death, removal of tibia. Specimens with implants of undecalcified sections, bone histomorphometry observation line.Results:1. All implanted implant were stable, not loose, the implant site wound healing.2. Bone measurement OUTCOME MEASURES:The combination of the width of the bone plate, the difference between the diameter of trabecular bone, cancellous bone area of the implant-bone contact ratio and bone mass of cancellous bone area, each group of indicators were statistically significant (P<0.05), pairwise comparisons found that in addition to the estrogen group and the simvastatin group{D1(p=0.85>0.05); D2(p=0.696>0.05); C (%)(P=.530>0.05); of V (%)(P=0.446>0.05)},the rest of the group differences were statistically significant.Conelusion:Experimental osteoporosis may hinder the implant and the bone tissue binding, the peri-implant trabecular bone reduction, combined with bone plate and bone trabeculae become thin, sequential therapy can effectively promote bone formation, thereby improving the bone tissue to the implant support.
