Objective: To detect and compare the expression of integrinα5β1 and fibronectin(FN) inosseous tissue around implant between Type 2 Diabetic Rats and normal rats, and explore thepossible mechanism that diabetic mellitus affect the implant osseointegration.Methods: Establish the rat model of type 2 diabetes,then 48 rats were divided into fourgroups randomly:normal implanting group (group C),normal control group(group C0),diabeticimplanting groups (group T),diabetic control group(group T0). The group C and group T wereimplanted the titanium implant near the epiphyseal end of tibia. Rats were killed after two,fourand eight weeks in batches. Using immunohistochemistry method the expression of integrinα5β1 and FN were detected in osseous tissue around the implants in each group.Result:(1) General observation: All experimental animals were no postoperative inflammation andno sudden death happended. Check up the stability of implant regulary ,the implant stabilizationof each stage were very well. The organization around the implant was no flare, and the colorwere very well also.(2) The results observed by microscope showed as follows: tibia cortex around the implantedplaces is thin which is partly absorbed, the width of ossis is increased, the texture of spongy boneis sparse and becoming thinner in the DM implanting group and the DM control group. Theperi-implant density of the DM implanting is lower and bone density of the last two groups isregular in The implanted bodies of the normal group in which ossis is narrower compared to thefirst two groups. the texture of spongy bone density is compact and even.(3) The expression of integrinα5β1 and FN:the expression in osseous tissue around theimplants in group T were significantly stronger than the expression in group C in two and fourweeks(P<0.05). Eight weeks after implantation, the expression of integrinα5β1 in group Twere close to that of group C, while the expression of FN in group T was significantly strongerthan group C (P<0.05).Conclusion:(1) Bone of Diabetic mellitus rats is thin which is partly absorbed, the width of ossis isincreased, the texture of spongy bone is sparse and becoming thinner. (2) Diabetic mellitus may weaken the implant osseointegration by changing expression ofintegrinα5β1 and FN in osseous tissue around the implants.
