Objective Establish rat70%hepatic ischemia-reperfusion model, injecting propofol to postconditioning on the model, observe the change of histomorphology,the expression of Bc1-2and Bax protein and the apoptosis index (AI) of hepatic cells, and investigate the effects of propofol postconditioning intervention on the ischemia reperfusion injury (IR) in rat livers.Methods Sixty Wistar rats,weighting200-250g,were randomly divided into5groups(n=12):(1)sham operation group(Group S);(2)ischemia-reperfusion group(Group IR);(3) postconditioning group with lmg/kg propofol (Group P1);(4)postconditioning group with2mg/kg propofol(Group P2);(5)postconditioning group with4mg/kg propofol(Group P4).Rat modeles of hepatic ischemia-reperfusion were established,and45mins reperfusion, Group P1, Group P2and Group P4were using physiological saline to propofol diluted to2ml, in5minutes before reperfusion via the femoral vein injecting1,2,4mg/kg propofol respectively. Group S and Group IR infusion of an equal volume (2ml)of physiological saline solution. The rats were killed respectively at3hours after reperfusion. Histomorphologic changes of the livers were assessed by HE method, the expression of Bcl-2and Bax protein of the livers was estimated by immunohistochemical staining method,the apoptosis index (AI) of hepatic cells was estimated by TUNEL method.Results1. The results of Light microscopy:To compare with Group S, the pathological lesion were more serious in Group IR, Group P1, Group P2and Group P4.To compare with Group IR,the histomorphologic changes were lighter in Group P1, Group P2and Group P4.2. The expression of Bcl-2and Bax protein:The expression of Bcl-2and Bax protein of the livers were extremely higher in Group IR, Group P1, Group P2and Group P4than in Group S,the difference between groups is statistically significant (P<0.05). The expression of Bcl-2protein was significantly higher in Group P1, Group P2and Group P4than in Group IR,while the expression of Bax protein were significantly lower than Group IR (P<0.05).Compared with Group P4, the expression of Bcl-2protein in Group P1and in Group P2were obviously higher,the expression of Bax protein were obviously lower (P<0.05). The difference between Group P1and Group P2has not statistical significance (P>0.05).3. The results of AI:AI were significantly higher in Group IR, Group P1, Group P2and Group P4than in Group S,the difference between groups is statistically significant (P<0.05). To compare with Group IR,AI were significantly lower (P<0.05) in Group P1, Group P2and Group P4.Compared with P4group, AI were lower (P<0.05) in Group P1and Group P2. The difference between Group P1and Group P2has not statistical significance (P>0.05).Conclusions1. Propofol postconditioning can increase Bcl-2protein expression and reduce Bax protein expression, propofol postconditioning on ischemia reperfusion induced liver injury maybe has a protective effect on the cellular apoptosis.2. The clinical dose propofol postconditioning can relieve the liver cellular apoptosis.
