Background: In clinical, analgesic method is often used to reduce the pain of patient withfracture or postoperative treatment.non-steroidal anti-inflammatory drugs (NSAID) is acommonly used drug.But in recent years,it can be found from most scholars’ study that NSAIDanalgesic at the same time will slow down the speed of fracture healing.With the improvement ofthe structure of analgesic drugs,a class of non-steroidal drugs carrying nitric oxide (NO),nitricoxide donor body non-steroidal anti-inflammatory drugs (NO-NSAID) was synthesized.Thesedrugs not only have anti-inflammatory and analgesic effect the same as traditional NSAID,alsocan protect the gastric mucosa to reduce adverse drug reactions due to the release of the NOmoiety.In addition,the role of anti-osteoporosis has been found in the latest study.Thus,the studyon mechanisms of these two drugs becomes the focus of attention.Objective: Under observation of the role of the traditional non-steroidal anti-inflammatorydrugs (flurbiprofen axetil) and new drug nitric oxide donating non-steroidal anti-inflammatorydrugs (NO-flurbiprofen axetil) on fracture healing in mice fracture model,this study aimed toexplore the effect of the two drugs on fracture healing treatment,providing a new way for thepain treatment in clinical fractures.Methods:45male BALB/c mice,with the similar body weight,4months age,were dividedinto3groups and modeled the fibula fracture separately.Then,different drugs(placebo,NO-flurbiprofen axetil and flurbiprofen ester) were injected at the same dose.Limb ofthe gross specimen and imaging X-ray observation were took after surgery two and fourweeks,measuring fracture bone mineral density and skeletal muscle HE staining histological testafter surgery4weeks.The SPSS16.0software package was used for statistical analysis.Results: It can be found that NO-flurbiprofen group was better than the other two groups byLimb of the gross specimen and imaging X-ray observation after4weeks,the bone mineraldensity at the fracture of NO-flurbiprofen group was higher than the other two groups by bonemineral density test,and the deference is statistically significant(P <0.001).By skeletal muscleHE staining histological test, the volume of the limb skeletal muscle cells and muscle fiber ofNO-flurbiprofen group was better than the other two groups.Conclusions: Using the NO-NSAID not only can reduce the adverse reaction.To a certainextent,it can promote fracture healing of the mice at the same time.
