Prostate cancer is a common cancer that develops in the male reproductive system. Many factors, including environment, genetics, age, androgen, and diet, have been implicated in the development of prostate cancer. Xenotropic murine leukemia virus-related virus (XMRV) is the first authentic gamma retrovirus that can infect humans; it was originally identified in prostate cancer patients in2006. Pilot studies indicated that sporadic prostate cancer correlates with the RNase L R462Q variant in Caucasian. Other studies showed a strong link between sporadic prostate cancer and XMRV infection. In this study, we detected XMRV in Chinese normal people and sporadic prostate cancer patients, got the RNase L genotype information, and investigated the association of RNase L with sporadic prostate cancer.Methods:In this study, we collected156sporadic prostate cancer samples and392blood donor samples for XMRV detection and RNase L genotype analysis.1. RNase L genotype was determined by PCR from125prostate cancer patients and240normal blood donors.2. XMRV was screened by nested-PCR, western blot and Q-PCR from156sporadic prostate cancer samples and392blood donor samples. Results:1. According to our study the frequency of RNase L R462Q variant was implicated in up to17.2%of prostate cancer cases whereas with a similar frequency of20.2%in Chinese normal people.2. We detected16XMRV positive samples (5prostate cancer samples and11blood donors) in156prostate cancer patients and392blood donors by nested-PCR. But there was no western blot signal in all XMRV PCR positive samples. The viral sequences amplified from different samples were not entirely consistent, and all of them were mouse endogenous retrovirus (mERVs) through the Blast alignment tools.3. The Q-PCR results showed that the virus copy was very low, range from0.2-8.4copies/μl plasma.Conclusions:R462Q variant was20.2%in Chinese, compared to35%in Caucasian populations. Statistical analysis showed that RNase L R462Q genotype and Chinese sporadic prostate cancer had no significant correlation.XMRV and related virus could be detected in3.2%prostate cancer patients and2.8%blood donors in China. Our study showed that there was no association of XMRV with prostate cancer in Chinese. Different types of mERVs were detected, which suggested that there may be different virus infection in Chinese.
