Objective: To investigate AQP2expression in renal collecting ducts inrats after renal ischemia reperfusion injury and the effects of sevofluranepretreatment on renal ischemia reperfusion injury.Methods: Twenty-seven adult male Sprague-Dawley rats were randomlydivided into three groups(n=9).B group: Performed a similar operationwithout occlusion of renal blood flow. I/R group: Performed the operation ofrenal ischemia reperfusion(I/R). Pre-s group: inhalation of2.2%(1MAC)sevoflurane for60minute were performed before I/R. After45minutes o fischemia and24h of reperfusion, serum BUN, Cr, SOD,MDA levels weremeasured. The kidney slices were observed under microscope and renalpathological changes were recorded, the tubular damage was scored. Collecturine to observe the change of the urine volume and the urine specificgravity.The levels of AQP2in renal inner medulla was detected by immuno-histochemistry and Real time PT-PCR. Comprehend the degree of renal injury.Results: after reperfusion, the serum BUN, Cr,MDA level in Pre-s groupand I/R group were significantly higher than those in B group (P<0.05); theserum BUN, Cr,MDA levels S group were significantly lower than those in I/Rgroup after reperfusion24h(P<0.05). the serum SOD level in Pre-s group andI/R group were significantly lower than those in B group (P<0.05); The SODlevels Pre-s group were higher than those in I/R group; Compare with B group,the urine volume were higher than those in I/R group and Pre-s group,and theurine specific gravity were higher than those in I/R group and Pre-s group.Theresults of the light microscope:nephric tuble were destroyed seriously, interstitial substance congestion and hemorrhage, large number ofphlegmonosia cella are present in the kidneys of rats in group I/R,but thesechanges are attenuated in group Pre-s. AQP2expression were significantlydecreased in Pre-s group and I/R group, Pre-s group were higher than those inI/R group.Conclusion:(1)AQP2expression in renal collecting ducts were significantly decreasedafter45minutes o f ischemia and24h of reperfusion.(2)Pretreatment with2.2%sevoflurane for60minutes can providesignificant protection against renal ischemia-reperfusion injury.(3)Sevoflurane up-regulating the expression of AQP2in ischemiareperfusion.(4).The mechanism of isehemic preconditioning against renal IRI maybeassociated to up-regulating AQP2Mrna and protein expression.
