1The effect of septic rat on myocardial functionObjective:To investigate the changes of sepsis on myocardial function in the isolated rat heart model. Method:Twelve male SD rats were randomized into sham group and sepsis group, in which the sepsis model adopts cecal legation perforation method (CLP). After operation at12h, the concentration of biochemical index (CK-MB, CK, ALT and AST) were detected to define the sepsis model. The isolated rat heart was perfused in a Langendorff apparatus after operation at12h. Heart rate(HR), left ventricular developed pressure(ΔLVP),+dp/dtmax,-dp/dtmax, and coronary perfusion flow(CPF)were measured in perfusion at10min,20min,30min,40min,50min,60min. Results: compared with sham group, myocardial functional parameters (ΔLVP,+dp/dtmax,-dp/dtmax) and coronary flow (ml/min) in sepsis rats were significantly (P<0.05) reduced at each points, but no significantly change was observed in the HR(P>0.05).Conclusions:myocardial injury incurred in early stage of sepsis in isolated rats heart, mainly myocardial systolic and diastolic functions are depressed.2The Expression of Glucose-regulated Protein78in Myocardial Injury of Sepsis in RatsObjective: To investigate the expression of glucose regulation protein78(GRP78) in heart tissue of the sepsis rat. Method:Twenty-four male SD rats were randomized into sham group and sepsis group(0h,4h,8h,12h,24h), in which the sepsis model adopts cecal legation perforation method (CLP). The changes of blood pressure (BP), heart rate (HR) and electrocardiogram were determined, the concentration of myocardial enzymes (LDH, α-HBDH, CK-MB and CK) were detected. Morphological changes of hearts tissues were observed by light microscopy. The expression of GRP78in heart tissue was detected by Quantitative fluorescence polymerase Chain reaction (real-time PCR). Result: compared with sham group, the BP in sepsis rats decreased significantly. Oppositely, the HR in sepsis rats increased significantly. ST depression was observed along with the significantly increase of myocardial enzyme. Importantly, it is shown that the expression of GRP78in heart tissue of CLP rats was significantly increased (P<0.05). Conclusion: The over-stress response of endoplasmic reticulum in sepsis probably results in myocardial injury.
