| Objective:It is to discuss the expressing situations and differences of CyclinD1and β-catenin in benign parathyroid tissue and sporadic parathyroid adenomas, and their relationship with clinical significance, which can make certain experimental base for clarifing the role of Wnt/β-catenin signaling pathway in the occurrence of sporadic parathyroid tumor.Methods:Altogether38pieces of surgical specimen (with20cases of parathyroid adenomasã€10cases of hyperplastic tissues and8cases of normal parathyroid) were collected between2006~2011, Shanghai Sixth People’s Hospital. All the specimen were confirmed by pathology, normal parathyroid glandular tissue came from the suspicious lymph nodes in thyroid cancer radical mastectomy, and all the patients had detailed clinical data. Fluorescent quantitation PCR was adopted to test mRNA expression of CyclinD1and β-catenin in parathyroid adenoma and normal parathyroid, using immunohistochemistry to detect the expression of Cyclin D1and β-catenin protein in sporadic parathyroid gland tumor and normal parathyroid tissue at the same time. After datas collected, statistics method was adopted to analyze expression difference of two kinds of genes and proteins in parathyroid adenoma and normal parathyroid tissue, and the relationship with clinical significance.Results:Fluorescent quantitation PCR analysis showed that:the expression of CyclinD1mRNA in adenomas and normal parathyroid tissue were2.36±1.12and1.50±1.03(P<0.05), β-catenin mRNA expression in adenomas and normal parathyroid tissue were1.02±0.45and0.88±0.56(P>0.05). However, the expression of CyclinD1and β-catenin mRNA have no obvious correlation with patient age, tumor size and serum calcium level in sporadic parathyroid tumors.The results of immunohistochemical showed that:the expression of CyclinDl protein was significantly higher in parathyroid adenoma and proliferation group than the normal parathyroid group (P<0.05), but there was no significant difference between adenoma group and proliferation group (P>0.05) the expression of β-catenin in cell membrane is downward with normal parathyroid tissueã€parathyroid gland hyperplasia and sporadic parathyroid tumor expression, but gradually increased in the cytoplasm and the nucleus; The abnormally expression of β-catenin was significantly correlated with the overexpression of CyclinDl expression in sporadic parathyroid tumors (P<0.05).Conclusions:1.There is abnormal expression of β-catenin, which is accompanied with the conversion process of normal parathyroid from hyperllasia to adenoma.2. The abnormal accumulation of β-catenin in the nucleus of parathyroid adenoma may start the transcription of CyclinD1gene, thus contributing to parathyroid cell proliferation and differentiation. |
