Objective:To determine the location,phenotype and quantity of decidual and peripheral bloodnatural killer cells in patients with unexplainable recurrent spontaneous abortions (URSA),evaluate the natural killer in peripheral blood after curettage of uterus in six month,investigate the relationship between the pathogenesis of URSA and differentiation of NKcells in human gestation, and to Reveal the clinical meanings of detecting the quantity ofNK cells in blood.Methods:1. collect the decidua and peripheral blood before curettage of uterus in URSApatients and nomal pregnancy, preparate it into frozen sections, the location phenotype andquantity of NK cells was detected by immunofluorescence in decidua.And phenotype andnumbers of NK cells was detected by Flow cytometry in blood.2. Collect the peripheral blood of URSA patients and normal pregnancy aftercurettage of uterus in the first, second, third, forth, fifth, sixth month. phenotype andnumbers of NK cells were detected by Flow cytometry.Results:1. NK cells aggregate around arteries. CD56~+CD16~+NK cells’quantity are (8.82±4.2)/20HPF and (4.59±5.7)/20HPF in two groups, CD56~+CD16~+NK cells in URSA group aremore than that in control group significantly (P<0.05).Two groups of decidual NK cellsCD56~+CD16~+/CD56~+CD16~-are (0.25±0.13) and (0.11±0.08), URSA group is more thanthat in control group significantly (P<0.05).2. CD56~+CD16~+NK cells occupy most of the blood.The percentage of CD56~+CD16~+NK cells are (15.2±5.29)%in URSA group,(11.9±3.4)%in the control group. The percentage of CD56~+CD16~+NK cells in URSA group are higher than that in control groupsignificantly (P<0.05).3. The percentage of CD56~+CD16~+NK cells are (13.9±4.0)%、(14.5±5.2)%、(14.4±5.8)%、(13.0±4.5)%、(15.3±5.8)%and (14.2±6.1)%in URSA group after curettage,(10.8±5.4)%in the control group. When compared with the normal nonpregnant group, the ratio ofNK cells were significantly increased after aborted at1th,2nd,3rd,5th,6th month (P<0.05).Conclusion:1. CD56~+CD16~+NK cells’quantity in decidua are more in URSA patient than normalpregnancy. and CD56~+CD16~+NK cells are gathered aroud the blood vessel apparently,andthe high quantity is lead to vascular abnormalities in the progress of embryo implantationof maternal-fetal interface,and is related with the URSA.2. CD56~+CD16~+NK cells’quantity is more in URSA than normal pregency, and wediscover that CD56~+CD16~+NK cells’percentage is higher than12%afer abortion in longtime. The high quantity of CD56~+CD16~+NK cells are displace to maternal-fetal interface,tobe the reason of URSA. So CD56~+CD16~+NK cells’percentage beyond12%can be used todignose, heal and evaluate the therapeutic effect of URSA.
