| [Objective]To investigate the expression of Fascin protein in ovarian serouscystadenocarcinoma and its correlation with clinical pathological parametersand prognosis. To further explore its mechanism and role in ovariantumorigenesis and progession for improving early diagnosis of aggressivetumours.[Methods]Paraffin sections using tissue microarrays including43case of ovaryserous cystadenocarcinoma,22cases of ovary borderline serous cystadenoma,17cases of Ovary serous cystadenoma and18cases of normal ovary tissuewere investigated by EnVision immunohistochemical method, the level ofexpression of Fascin, Ki-67and E-cadherin were evaluated in tissues and thecorrelation between their expression and the clinicopathological parameterswere analyzed statistically.[Results](1) Fascin expression was significantly enhanced in serouscystadenocarcinom(a35/43,81.4%)and borderline serous cystadenom(a9/22,40.9%)compared with benign serous cystadenom(a2/17,11.8%)P<0.01).There was increased expression of Fascin in adenocarcinoma compared withbordline tumors.(2) Fascin expression correlated well with tumor histological grade and lymph node metastases(P<0.01);and Fascin overexpression also wassignificantly associated with a poor prognosis(P<0.05).(3) E-cadherin expression positive rate in ovarian serouscystadenocarcinoma was55.81%(24/43) which was lower significantly thanborderline serous cystadenoma (90.91%,20/22) and benign serouscystadenoma(100%,18/18)(P<0.05), E-cadherin expression correlatedwell with lymph node metastases in ovarian serous cystadenocarcinom(aP<0.05).(4) Ki-67expression was significantly enhanced in serouscystadenocarcinoma(43/43,100%)compared with and borderline serouscystadenoma(13/22,59.1%)compared with benign serous cystadenoma(5.9%,1/1711.8%)(P<0.01),and Ki-67expression is associated withtumor histological grade (P<0.05); Ki-67overexpression also wassignificantly associated with a poor prognosis(P<0.05).(5) There was positive correlation between Fascin and Ki-67expressionin ovary serous cystadenocarcinoma(P<0.01), and expression of Fascinwas Negatively correlated with E-cadherin(P<0.01).[Conclusion](1)The over-expression of Fascin and Ki-67and down-regulation ofE-cadherin expressing could be early events during ovarian serouscystadenocarcinoma development process.(2) Fascin expression correlated well with ovarian serouscystadenocarcinoma histological grade and lymph node metastases; Ki-67expression is associated with tumor histological grade and E-cadherinexpression is associated with lymph node metastases in ovarian serouscystadenocarcinoma. (3) Overexpression od Fascin and Ki-67were significantly associatedwith a poor prognosis in ovarian serous cystadenocarcinoma; but there wasnot correlation between E-cadherin expressing and prognosis in ovarianserous cystadenocarcinoma.(4) There was positive correlation between Fascin and Ki-67expressing,and Fascin was correlated with E-cadherin negatively in ovarian serouscystadenocarcinoma. |
