Veratrum (family Liliaceae), refered to a genus of poisonous and perennial herb, is a commonly used traditional Chinese medicine (TCM) for the treatment of aphasia arising from apolexy, dysentery, jaundice, headache, scabies and chornic malaria. The modern research has showed that Veratrum alkaloids have great potential in cancer treatment. Thus the systematical study on extraction, separation, identification and activity of Veratrum alkaloids are worth carrying out.This paper established a fast and reliable method based on Near-infrared Spectroscopy (NIR), for the identification of Veratrum patulum, Veratrum nigrum L. and Veratrum nigrum L. Var. ussuriense Nakai. The2nd derivation with9point smoothing was utilized for spectra pretreatment and factorization method was conducted at the range of5150-5600cm-1and6750-7450cm-1. The obtained Selectivity (S value) were all above4. Unknown samples were classified using the established model, and none of them was identified as the wrong class.In this paper, the chemical constituents from rhizomes and roots of Veratrum patulum were studied. Finally,10compounds were isolated and purified by numerous separation technologies, such as, silica gel chromatography, preparative high performance liquid chromatography (pre-HPLC), centrifugal partition chromatography and thin-layer chromatography, etc. These compounds were unambiguously identified on the basis of MS、 IR、UV、1D-NMR(1H,13C-NMR),2D-NMR(1H-1H COSY、HSQC、HMBC), and they were (3-Sitosterol (1), β-Daucosterol (2), Dodecanoic acid glyceride (3),25Ene acid glyceride (4), Jervine (5), Veratramine (6), Presudojervine (7), Veratramin-3-O-a-D-glucopyranoside (8), Veratramin-23-O-α-D-glucopyranoside (9), Verdine (10).The cytotoxic effect of the isolated alkaloids (Compound5-10) against human pancreatic cancer ASPC-1and PANC-1cells were investigated via MTT assay. All the six steroidal alkaloids showed inhibitory activity. Among them, the IC50values of compound5and compound9on ASPC-1were202.10μmol·L-1and203.65μmol·L-1, respectively. The IC50values of compound5and compound6on PANC-1were3.50μmol·L-1and14.08μmol·L-1, respectively.
