Objective:To find the effective monomer components of Zhuanggu Zhitong prescription, and to explore the possibe mechanism.Methods:Eighty female SD rats weighing about250g were randomly divided into model group, Zhuanggu Zhitong prescription group, icariin group, oleanolic acid group, psoralen group, naringin group, icariin+oleanolic acid+psoralen group, and Sham operation group.Five days after ovariectomization, different treatment were lasted for13weeks.Blood and lumbar bone, tibia bone samples of the rats were used to determine the bone mineral density, bone biomechanics, path-ology and other aspects to evaluate the efficacy.Results:1.Icariin group and oleanolic acid group showed significantly increase in lumbar bone mineral density(P<0.05); could effectively prevent osteoporosis.2.Icariin group could obviously increase the E2level (P<0.05) in order to reduce the rate of bone turnover, reduce the loss of bone’s, in order to achieve the anti role in osteoporosis.3.Icariin+oleanolic acid+psoralen group could significantly enhance the trabecular bone density (P<0.05), could reverse osteoporosis with improved pathological evidences.4.Icariin group, oleanolic acid group, psoralen group, naringin group, icariin+oleanolic acid+psoralen group, and Sham operation group could do nothing to improve the castrated rat bone biomechanical(p>0.05).Conclusion:1.The icariin and oleanolic acid is the effective monomer composition.2.Icariin group could increase the level of estrogen, reduce bone turnover, inhibit the transformation of bone cells, reduce bone formation and inhibit bone resorption.3.oleanolic acid group can treat postmenopausal osteoporosis mechanism may be through regulation of the expression of matrix metalloproteinases to promote bone formation.4.Icariin group, oleanolic acid group, psoralen group, naringin group, icariin+oleanolic acid+psoralen group, and Sham operation group could increase the average trabecular bone density, reversal of bone tissue pathological change.5.Zhuanggu Zhitong prescription can treat postmenopausal osteoporosis mechanism may be through improving the level of estrogen and (or) playing a role like estrogen by the effective ingredients, regulation of the expression of matrix metalloproteinases, reduced the rate of bone turnover, made the osteoblast proliferation than osteoclast formation, reducd the osteolysis phenomenon to improve bone mineral density.
