The Experimental Study On The Effects Of Liuweidihuang Decoction On Col-â…¢ And Co-â…£ Expression With5/6Nephreetomy Rats

Objective:Observation of Liuweidihuang Decoction on5/6nephrec-tomized rat remnant kidney of collagen type III, collagen type IV expression influence, discussion of Liuweidihuang Decoction to prote-ct renal function, the mechanism of delaying kidney damage.Methods:In accordance with the completely random methods60male SD rats were divided into5groups:Black group, sham operation group, model group, enalapril group, Liuweidihuang Decoction group,12rats in each group. The model group, the enalapril group, Liuweidihuang Decoction group with5/6nephrectomy induced chronic renal failure rat model, Sham operation group undergoing operation two times, each time only open celiac, exposed kidneys, abdominal closure. Second operation after7days, the rats were treated by gavage. In eighth weeks in rats were sacrificed, observation of remnant kidney tissue morphological changes were observed with immunohistochemistry rat kidney Col-III and Col-IV expression.Results:1By using HE and Masson staining with5/6nephrec tomy rats renal tubular interstitial tissue morphological changes,5/6nephrectomy induced renal damage in rats model of successful re-production, Liuweidihuang Decoction and enalapril group can redu-ce renal tubulointerstitial damage and fibrosis degree;2.Immunohist-ochemical semiquantitative analysis revealed six rehmannia root gro-up of renal cortical Col-III and Col-IV expression is less than the model group;3. Liuweidihuang Decoction can lower blood levels of BUN, Scr,and reduce24hours urine protein quantity etc.Conclusion:Liuweidihuang Decoction may be adopted to decrease the accumulation of extracellular matrix, downregulation of Col-III, Col-IV expression, make the cell matrix components decreased synthesis, in order to reduce inflammation and fibrosis, to prevent renal interstitial fibrosis, protect the kidney function.
