Objective:To study the effects of Yang jing capsule on rat sexual function and the the expression of P-Erkl and P-Aktl kinase in penile cavernous tissue of aging rats, and analyse its possible mechanism by multiple analysis of animal experiments.Methods:All40aging male SD rats (20months old), were randomly divided into the control group and the experimental group averagely. The experimental group rats were given Yang jing capsule by gavage, at the dosage of0.5g/kg.d-1, and the control group was given at the same dosage. Four weeks later, we measured the total serum testosterone levels of rats in two groups, and recorded the data; Subsequently, killed all rats and isolated the the corpus cavernosum by surgical operation. Keeping a part of specimen by4%paraformaldehyde phosphate buffer storage, and testing the expression of CD34and eNOS with immunohistochemistry. The rest of tissue were preserved to70degrees below zero by liquid nitrogen for testing the expression of P-Erkl mRNA and P-Aktl mRNA with RT-PCR detection.Results:1.The control group of aged rats after intervention of total serum testosterone levels decline than those before intervention, and the comparison group showed no significant difference(P>0.05); In the Yang jing capsule group, the level of total serum testosterone rised up after intervention, but no significant difference(P>0.05);2.The penile erection, erection latency times and maintenance of erection time of experimental group rats improved than the control group, but no statistical difference(P<0.05);3.The expression of CD34and eNOS on the Yang jing capsule rats penile corpus cavernosum tissue were all significantly enhanced, showing a significant difference(P<0.05); Compared with the control group, the expression of P-Erkl mRNA in experimental group rats from penile corpus cavernosum tissue decreased obviously, with statistical difference(P<0.05), but the change of P-AktlmRNA in the comparison group showed no significant difference;4.The HE morphology staining results:penile corpus cavernosum tissue of aged rat in the experimental group showed that, the vascular lumen dilated, congested markably, and the inner wall got thinning, but there were no obvious change in the control group, and the lumen resided much old blood;5.Immunohistochemical results showed that, there were lots of positive deeply dyed brown granules in two groups of rat corpus cavernosum tissue, but the degree of dyed and particle density were not different. Observing at low magnification vision, the number and strength of eNOS positive granules expression in the experimental rats were significantly higher than those in the control group of rats;6.Microvessel density results:the expression of CD34in the experimental rats’cavernous vascular endothelial tissue was stronger than that of the control group, the average count of MVD in the experimental group increased compared control group.Conclusion:Yang jing capsule can improve the aging rats’sexual function and erectile function.We thought it was possiblely the comprehensive effects of the level of increasing total serum testosterone levels or delaying total serum testosterone levels decline, improving penile vascular circulation, inhibiting the activity of P-Erkl kinase in the penile cavernous.
