Effect Of Herbal Compound For Reinforcing Kidney,Activating Blood And Arousing Consciousness On The Expression Of Bcl-2and Bax Of Organ Of Corti In Gentamicin-induced Ototoxic Mice

ObjectiveTo observe the hearing protection of herbal compound for reinforcing kidney,activating blood and arousing consciousness(HCRAA) in gentamicin(GM)-induced ototoxic mice, analysising the expression of Bcl-2and Bax in the Organ of Corti in damaged internal ear and approaching correlated mechanism of multi-target action of traditional Chinese medicine.MethodsHerbal compound for reinforcing kidney,activating blood and arousing Consciousness (composition:Drynariae,epimedium,astragalus,Chinese angelica,Salvia,Chuanxiong,Acorus gramineus,leech, magnets,etc.) were decocted for high,medium and low concentrations of the three decoction containing respectively pharmacognostic2.375g/ml,0.475g/ml,0.095g/ml (based on the mouse body weight by25g,0.8ml of intragastric administration every day, the decoction of the concentration of one is equivalent to70kg adult day clinical routine the equivalent amount of volume, high concentration and low concentration of decoction were respectively equivalent to the amount of5times and0.2times the dose).40Kunming mice were randomly divided into5groups of12. A, B, C, D group:be respectively taken HCRAA high, medium and low concentration and normal saline by of every day,100mg/kg, for15consecutive days. From the first day of GM intraperitoneal injection, four groups be respectively intragastric administratered for high, medium and low concentration of Chinese compound decoction and normal saline, for20days; E groups:contrast group, normal feeding, not any treatment. After administration the mice in each group were measured auditory brainstem evoked potentials (ABR) thresholds, and adopted the method of immunohistochemistry to analysis the expression of B-cell lymphoma/Leukemia-2(Bcl-2) and Bcl-2-associated-X-protein (BAX) in the Organ of Corti.ResultsExpression of Bcl-2and Bax in the Organ of Corti of mice:HCRAA of high and middle concentration could elevate the expression of Bcl-2, compared in these two groups and with normal contrast group, the result had no significance; HCRAA of high and middle concentration could degrade the expression of Bax, compared in these two groups and with normal contrast group, the result had no significance.ConclusionHCRAA could effectually antagonize ototoxicity of GM and protect the Organ of Corti of mice. HCRAA could elevate the expression of Bcl-2and degrade the expression of Bax in the Organ of Corti, obviously improve hearing and play an important action in protection the function of the Organ of Corti when taking ototoxicity medicine meanwhile taking HCRAA medicine.
