[Objectives] To compare the flavonoids content of Allium cepa extracted from water and ethanol, and the influence of the extractions on colorectal cancer cell proliferation、apoptosis and cycle.[Method] Allium cepa was extracted by water and ethanol, the flavonoids content of the extractions were detected by UV. Colorectal cancer cells(Lovo cells、SW480cells、HT-29cells、 HCT-8cells) were cultured in vitro, the proliferation ability of cultured cells in varying concentration of flavonoids (0、2.5、5、10、20、40、60、80mg/L)were tested by CCK-8analysis. The apoptosis of cultured cells in varying concentration of flavonoids (40、60mg/L)were detected by flow cytometry, the influence on cell cycle was tested by concentration of flavonoids40mg/L.[Results] The content of flavonoids extracted by water was higher than ethanol. Both of the water extraction and ethanol extraction (tested by CCK-8analysis) inhibited the proliferation and showed a dose dependence and time dependence to colorectal cancer cells.The water extraction inhibited colorectal cancer cells proliferation was more significant than the ethanol extraction. Both of the water extraction and ethanol extraction (detected by flow cytometry) could enhance the colorectal cancer cells’apoptosis rate and showed a dose dependence, the apoptosis rate induced by water extraction was higher than ethanol extraction in SW480and HCT-8cells, but the ethanol extraction caused a higher apoptosis rate than water extraction in HT-29cells. Allium cepa extractions could block the cell cycle at G1phase, hindered cells conversion to S phase then inhibited cell proliferation.[Conclusion] Allium cepa extractions could significantly inhibit the proliferation of colorectal cancer cells, also induced the apoptosis of colorectal cancer cells and showed a notable influence in cell cycle. The mechanism needs to be further study.
