Objectives: To explore the effects of celecoxib on cell apoptosis in human colorectal cancer cell line HT-29 and the expressions of MIF , PTEN and Caspase-3 .Methods: MTT assay, Flow cytometry (FCM) and AO-EB staining were used to quantify the influences of different concentrations of celecoxib on the apoptosis in colorectal cancer cell line HT-29 .The expressions of MIF, PTEN and Caspase-3 were observed by RT-PCR and Western blot analysis.Results: 1. The results of MTT assay showed that HT-29 cells survivals rates decreased with the increase of celecoxib concentrations from 0 to 100μM after treated with different concentrations of celecoxib for different periods; 2. The results of FCM showed that the proportion of HT-29 cells increased in G0/G1 phase and G2/M phase and decreased in S phase with the increase of celecoxib concentrations from 0 to 100μM when HT-29 cells were treated with different concentrations of celecoxib for 24 hours;3.The results of AO-EB staining showed that celecoxib induced the apoptosis of HT-29 cells; apoptosis bodies and apoptotic index were increased with the increase of celecoxib concentrations from 0 to 100μM; 4. The results of RT-PCR analysis showed that the expressions of MIF mRNA in HT-29 cells were down-regulated with the increase of celecoxib concentrations from 0 to 100μM after HT-29 cells were treated with different concentrations of celecoxib; 5. The results of Western blot analysis showed that the expression of MIF protein in HT-29 cells was down-regulated, whereas the expressions of PTEN and Caspase-3 proteins in HT-29 cells were up-regulated with the increase of celecoxib concentrations from 0 to 100μM.Conclusions: 1.Celecoxib inhibits HT-29 cells proliferation and induces HT-29 cells apoptosis; 2.The mechanisms of the roles of celecoxib may be associated with down-regulation of the expression of MIF and up-regulation of the expressions of PTEN and Caspase-3.
