Objeetive: Comparative study on the effects and mechanisms of BushenHuoxueRecipe and YiqiBuxue Recipe for the Myelosuppression after Chemotherapy.Methods:160ICR mice, were randomly divided into4groups:the blank control,themodel control, the BushenHuoxue group, and the YiqiBuxue group,the latter threegroups of mice were injected cyclophosphamide intraperitoneally for3consecutive daysat100mg/kg daily on day8,9,10. The BushenHuoxue group of mice and the YiqiBuxuegroup of mice were administrated by gavage with BushenHuoxue decoction andYiqiBuxue decoction respectively for16days at0.3ml/20g/d,the same volume ofphysiological saline was given to the other two groups respectively. Thereafter,10micewere randomly taken for detecting the peripheral blood on0,1,4,7,11,14,17,21daysafter model established;For the rest30mice,10mice were randomly taken respectivelyon the1,3,7thday after modeling, the levels of the GM-CSF in the blood, the marrowapoptosis, BMNC,hematopoietic stem/progenitor cell CD34+,bone marrow cellcycle,thymus index and spleen index,and the pathology of the bone marrow tissue weredetected.Result:1. The model of myelosuppression was successfully established by intraperitonealinjection of cyclophosphamide100mg kg-1d-1,3consecutive days.2. The WBC count of the peripheral blood of the two herbal groups was obviouslyhigher than the model group at the lowest point which was on the1stday after themodel was established, YiqiBuxue group was higher than both the model group and theBushenHuoxue group at the peak which was on the7thafter modeling(P<0.05orP<0.01).The valley bottom of the two herbal groups of the PLT count of theperipheral blood was higher than the model group(P<0.05)and showed a peak on the 7thday after modeling,which was higher than the blank group(P<0.05). BushenHuoxuegroup was higher than YiqiBuxue group both at the lowest point and the peak,butdidn’t show obvious significance.3. The BMNC of the model group descended to the lowest point on the1stday, the twoherbal groups descended to the lowest point on the3rdday,and the extent of descentseemed to be lighter for the BushenHuoxue group and heavier for the YiqiBuxuegroup,which was heavier than the model group and turned up a similar peak as themodel group did on the7thday,but the peak was lower(P<0.05),while the peak ofBushenHuoxue group was not obvious,which was higher than the blank group,butdidn’t have statistical significance.4.Bone marrow cell cycle bone marrow cells of the model group were more in theG1phase(P<0.05)and less in the S phase(P<0.01)than the blank group.The changes oftwo herbal groups were similar to the model group,but bone marrow cells of theBushenHuoxue group were less in the G1phase and more in the S phase than themodel group and the YiqiBuxue group(P <0.05or P<0.01).5. Expression of bone marrow hematopoietic stem cells CD34+The expression ofbone marrow hematopoietic stem cells CD34+of the model group decreasedsignificantly compared with the blank group(P<0.01).The two herbal groups seemedlight compared with the model group and the YiqiBuxue group was slightly higher thanthe BushenHuoxue group,but there was no statistical significance.6.Apoptosis of bone marrow cells The ratio of early and late apoptosis of modelgroup and two herbal groups were higher than the blank group and the difference wasvery significant(P<0.01); And that of BushenHuoxue group seemed to be lower thanmodel group and YiqiBuxue group,but there was no statistical significance.7.Expression of cytokine GM-CSF in the serum The two herbal groups were higherthan the model group on the1stday after modeling(P <0.05or P<0.01). The extent ofdecline of the two herbal groups seemed light compared with the model group on the3rdand7thday after modeling,but there was no statistical significance.8.Bone marrow biopsy The shape of erythrocyte series, granulocyte series and macrophage series of the blank group was regular and well-distributed. Cells of allseries of model group, BushenHuoxue group and YiqiBuxue group reducedproportionately and obviously on the1stday after modeling; on the3rdday aftermodeling, cells of all series of model group still reduced proportionately and obviously,BushenHuoxue group and YiqiBuxue group reduced proportionately and slightly, butincreased compared with the1stday; Cells of all series of model group had a moderatereduction proportionately on the7thday,but recovered a bit compared with the3rdday,BushenHuoxue group increased compared with the3rdday, while that of YiqiBuxuegroup was regular and well-distributed.Conclusion:1.Intraperitoneal injection of cyclophosphamide100mg kg-1d-1,3consecutive days,you can successfully replicate the myelosuppression model after chemotherapy,consistent with the bone marrow suppression state of clinical malignancy in patientsreceiving chemotherapy.2. Two herbal prescriptions may lessen the reduction of white blood cells and platelets,speeded up recovery, and the YiqiBuxue method was better in reducing leukopenia,Bushenhuoxue method was better in preventing and curing thrombocytopenia.3. Bushenhuoxue decotion could relieve the inhibition of G1phase after chemotherapy,and promoted the proliferation of hematopoietic cells and recovered BMNC steadily;while YiqiBuxue decotion could raise the number of bone marrow hematopoieticstem cells and had favourable effect in recovering BMNC and the cells oferythrocyte series, granulocyte series and macrophage series.The pathways,links andspecific efficacy of the two herbal groups may be different on the prevention andtreatment of myelosuppression after chemotherapy.
