Prostate specific antigen is a glycoprotein secreted by cells of the prostate gland. The serum PSA has been recognized as one of the important tumor marker for prostate cancer diagnosis. Based on a review of evidence and concludes that prostate specific antigen screening could result in reduction of mortality and death rate, prolong the life time of patients, and increase the subsequent treatments positively.Herein, two immunoassay methods based on sandwich reaction format were established for detection the concentration of PSA in human serum by using GoldMag(?) particles as the carriers. One is the electrochemiluminescence immunoassay (ECLIA) based GoldMag(?) microparticles and the other is lateral flow immunoassay (LFIA) based on GoldMag(?) nanoparticles.The GoldMag(?)-AS microparticles were used to conjugate the monoclonal anti-PSA antibody and the coupling condition was optimized. At the same time, the method for another anti-PSA antibody labeled with Ru(bpy)32+was also established and evaluated. PSA in human serum was reacted with the Ab on the surface of GoldMag(?) and the Ab labled with Ru (bpy)32+to form a sandwich complex. Finally, the quantitative detection of PSA in human serum was achieved through the electrochemiluminescence detection systems.GoldMag(/)-CS nanoparticles were firstly modified with PSS to obtain monodispersed colloid. The modified GoldMag NPs were used to label the monoclonal anti-PSA antibody and then LFIA based on GoldMag(?) was established for quantitative detection of PSA in human serum. The sensitivity, specificity, accuracy and stability were evaluated respectively. The results showed that the LFIA based on GoldMag(?) is an ideal point-of-care test which has the advantages of rapid, better quantitative and longer signal life time.
