Objective:To observe the wound healing with platelet-rich plasma (PRP) gel andadipose-derived stem cell(ADSCs) applied onto the wound; Explore the role ofADSCs and PRP in soft tissue defect repairing.Methods:Harvest adipose tissue from inguinalis fat pad of SD rats,isolation,culture,and identification the ADSCs through three differentiation method. Take30male SDrats about6-7weeks old randomly.Selected12of rats been take blood from heart.Preparation the PRP with modified Appel method.To count platelet of whole bloodand PRP under microscope.Take the remaining rats.Formed a skin defect wound byresection circular of2cm diameter both sides of skin apart from the center of thespine.Divided the rat into3groups (n=6) randomly, of which group A treatmentwith ADSCs and PRP; Group B treatment with ADSCs; Group C treatment whitPRP.Selected one side of skin defect wound for test Randomly and the relative sideskin defect is for controll, Handle all of the control wound to group D; Observe thegrowth of granulation tissue on the wound surface;observe the inflammatorysurrounding and the degree about epithelial. statistical analysis and Record thewound size, and calculation the shrinkage rate of wound in different periods,.Recordthe time of completely healing time.Histologic observation of the wound healingtissue.Results:Platelet counting showed platelet of PRP is5.21times than wholeblood. The wound completely healed time:Group A18.25±1.44days; GroupB19.13±1.28days;Group C19.72±0.87days; Group D22.31±1.65days, The timeof each treatment group and control group was significantly obvious (P <0.05).At3,7,11and15days after experimental treatment, compared with the control groupthe experiment group of wound contraction rate was significantly obvious (P <0.05);compared wound contraction rates between the experimental groups:Group A havethe highest of wound contraction rate than other groups was significantly obvious (P<0.05), and the wound contraction rate Between group B and group C difference were not statistically significant (P>0.05). Histological observation showed that:group A have a layered structures subcutaneous gland-like and neovascularizationcompared with normal skin tissue,In the Group B there have similar collagen fibersto normal skin tissue; gland-like structures wounds than normal skin.The PRP treatedgroup have more new blood capillaries and less gland-like structure, but the collagenfibers arranged are in order; In the control group the field of vision have lots ofdisorders collagen fibers.Conclusion:Application of ADSCs with PRP can enhance the quality and shortenthe wound healing time than used them alone.
