Objective:To investigate the effects of Dihydroartemisinin Onapoptosis of prostate cancer PC-3Cells by low-frequency ultrasound andexplore its mechanism.Methods:Prostate cancer cells PC-3was selected and were divided intofour groups. They were contral group, Dihydroartemisiningroup(50μmol/L、100μmol/L、200μmol/L), low-frequency ultrasoundgroup, and low-frequency ultrasound combined with Dihydroartemisiningroup(50μmol/L、100μmol/L、200μmol/L). Cell apoptosis of everygroup was analyzed by flow cytometry after48; Transmission electronmicroscope and scanning electron microscope was used to observe cellmorphous; Cell immunofluorescence was used to observe the changes offluorescence intensity among the different groups;Western blot was used toanalyze the expression of Bax and Bcl-2.Results:Flow cytometry results showed that the rate of apoptosis ofprostate cancer PC-3cells have no difference between control group andultrasound group,but there were significant differences between every low-frequency ultrasound combined with Dihydroartemisinin group andcontral group、low-frequency ultrasound group、Dihydroartemisiningroup, which were statistically significant differences(P<0.05).Typicalmorphology of apoptotic cells have observed between DHA group andultrasound combined with DHA group through Transmission electronmicroscope and scanning electron microscope.The fluorescence intensitywhich reflect the expression of Bax was significantly higher in ultrasoundcombined with DHA group than DHA group、control group and ultrasoundgroup,but the fluorescence intensity of Bcl-2in ultrasound combined withDHA group was weaker than DHA group、control group and ultrasoundgroup.According to the results of Western Blot:Compared with controlgroup、ultrasound group and DHA group,the expression of Bax wereobviously increased in ultrasound combined with DHA group,there wasstatistically significant differences between DHA group、controlgroup、ultrasound group and ultrasound combined with DHA group(P<0.05);the expression of Bcl-2were obviously decreased in ultrasoundcombined with DHA group,there were statistically significant differencesbetween control group、ultrasound group、DHA group and ultrasoundcombined with DHA group (P<0.05).Conclusion:Low-frequency ultrasound group combined with DHA canenhance apoptosis of prostate cancer PC-3cells by increasing the expressionof Bax and decreasing the expression of Bcl-2which might increase the permeability of cell membrane and enhance the medicine density of DHA incell.With increasing the concentration of DHA,low-frequency ultrasoundgroup combined with DHA can enhance apoptosis of prostate cancer PC-3cells,suggesting that dihydroartemisinin induce apoptosis of prostate cancerPC-3cells by low-frequency ultrasound may be related to the doseconcentration of dihydroartemisinin.
