Object To prepare paclitaxel-carrying nanometre microbubbles(PNM) modified with human serum albumin(HSA) and to assay of paclitaxel in micro bubbles by Reversed phase high-performance liquid chromatography (RP-HPLC).Method To choose lipid as Microbubbles forming materials,the HSA modified PNM was prepared by thin-film dispersion method and mechanical vibration.To determine the paclitaxel standard curve by RP-HPLC and study the stability,the degree of precision,and the recovery rate.Result The HSA modified PNM appearance is milky white,no flocculent,granular precipitate. The paclitaxel standard curve was A=28.734C+4.3239(r=0.9982).There was a good linear relationship within the concentration range of0.5μg/ml-7.5μg/ml of paclitaxel.The stability,the degree of precision,and the recovery rate were all fit the technology request.Conclusion The preparation technique was well-repeatable.The HSA modified PNM has uniform appearance.The way of determine content of paclitaxel in microbubbles is reliable. Object To prepare paclitaxel-carrying nanometre microbubbles(PNM) modified with human serum albumin(HSA) and study its characteristics and acute toxicity test in SD rats.Method The appearance,concentration,mean diameter,zeta potential and the entrapment efficiency were observed and detecte.It was stored at-4℃refrigerator for4weeks, to investigate its stability.The normal rabbit’s iliac artery contrast imaging was observed.The acute toxicity was experimented on SD rats and serum biochemical was analyzed.Result This modified PNM had a good shape.The mean diameter was(772.9±6.2)nm, the concentration of bubble was (8.64±1.38)×109/ml, zeta potenital was(-13.2±0.3)mv, the entrapment efficiency of the modified bubble was (93.51±2.07)%, which was higher than that of no modified bubble (84.88±4.14)%(P<0.05).It could make the rabbit iliac artery enhancement.The acute toxicity results indicated that there was no negative effect on serum biochemical in SD rats.Conclusion The HSA modified PNM was stable and effective, with smaller size and higher encapsulation efficiency.It can achieve enhanced artery enhancement.By means of HSA modified PNM1μl/g intravenous injection, the acute toxicity show that there was no negative effect on serum biochemical in SD rats.
