Objective:The purpose of this study was to investigate the association betweenADAMTS-1and acute CVS after experimental SAH in rats through testingthe expression of ADAMTS-1on basilar artery, discussing the role ofADAMTS-1in early brain injury after SAH.Methods:Total108healthy male adult SD rats (250-300g, weight) were selectedand randomly divided into2groups, sham-operation group and operationgroup. Autologous arterial blood in operation group rats were extracted,equivalent physiological saline were injected in sham group rats, toestablish experimental SAH model. The expression of ADAMTS-1onbasilar artery in different time points after SAH, such as6h,12h,24h,48h,72h, were detected by applying Western-Blot and RT-PCR. Meanwhile, themorphological change of basilar artery in rats was observed by HEstaining. Data was analyzed by one-way analysis of variance with SPSS 13.0.Results:Comparing with sham group, the expression of the total mRNA andprotein of ADAMTS-1have no statistical significance at6h, howeverincreased dramatically at12h after SAH and reached the peek at24h, afterthat decreased gradually at48h and72h in basilar artery. However, it werestill higher at48h and72h after SAH than that of sham grop. Meanwhile,we can also observed the morphological change of basilar artery by lightmicrosocopy which showed that the changes of basilar artery areinapparent at6h after SAH in rats, and then the calibrated diameter beganto shrink and the wall thickness increased from12h after SAH and theacute CVS occurred most obviously at24h, relieved at48h. Thecalibrated diameter and the wall thickness magnified much more distinctat72h compared to24h and48h after SAH. The expression of totalmRNA and protein of ADAMTS-1was positively associated with acuteCVS after SAH(r=0.916,p=0.003).Conclution:Our results suggested that there is a positive correlationship betweenADAMTS-1and acute CVS after SAH which maybe were involved in thepathological process of EBI after SAH in experimental rats.
