Objective: To observe the relation between the expression ofADAMTS-1in the hippocampus of rat and the permeability of blood-brainbarrier. Investigate the effect of ADAMTS-1in the early brain injury (EBI)after subarachnoid hemorrhage (SAH).Method: Totally108adult male SD rats were randomly divided into2groups, the sham-operation group and SAH group. The SAH model wasinduced by injecting300μl of autologous arterial blood into thesubarachnoid space near the circle of Willis via a cannula in an artificial holebetween the olfactory bulb and frontal lobe. The expression Of ADAMTS-1in the hippocampus were examined at6,12,24,48,72h by western-blottingand RT-PCR the BBB permeability was evaluated by detection ofextravasated Evans Blue.Result:1.The contents of Evans blue in rat brains at6and12h following SAH compared to sham group were unchanged.(P>0.05);They increaseddramatically at24h after SAH and Maintain a high level to72h.(P<0.05VSshame group)2. Compared to the sham group the level of ADAMTS-1mRNAexpression after SAH6and12h showed no significantdifference.(P>0.05):mRNA levels of ADAMTS-1were significantlyincreased at24h after SAH compared to sham-operated animals(P<0.05):Reached the peak at48h and Maintain a high level to72h.(P<0.05VS shamegroup).3. The contents of Evans blue in rat brains at6and12h following SAHcompared to sham group were unchanged.(P>0.05);They increaseddramatically at24h after SAH and Maintain a high level to72h.(P<0.05VSshame group)4. The protein expression of ADAMTS-1and the content of brain EBare positive correlated (r=0.936P=0.014)Conclusion: ADAMTS-1in the hippocampus of rats after SAH maybe involved in the pathological progression of early brain injury.
