Objective: To investigate and compare the effect of neuropilin-1(NRP-1) antisense oligodeoxynucleotides (ASODN) with that of vascularendothelial growth factor receptor-2(VEGFR-2) ASODN on theproliferation and apoptosis of human gastric cancer cells.Methods: Transmit thiophosphorylate modified antisense oligodeo-xynucleotides of NRP-1and VEGFR-2into human gastric cancer cellsSGC7901with liposome separately and simultaneously, detect NRP-1mRNA levels and VEGFR-2mRNA levels by reversetranscriptase-polymerase chain reaction (RT-PCR); Measure the changes ofcell proliferation by methyl thiazolyl tetrazolium (MTT); Observe cellapoptosis by flow cytometry.Results: The transcription level of NRP-1mRNA and VEGFR-2mRNA in SGC7901cells after transmission with ASODN decreased. NRP-1ASODN and VEGFR-2ASODN inhibit the proliferation of SGC7901cells and meanwhile promotes the apoptosis of these cells. Such effect intensifiesas the concentration of ASODN increases; When NRP-1ASODN andVEGFR-2ASODN are used separately, no significant difference is detected.Conclusion: NRP-1ASODN and VEGFR-2ASODN inhibited thetranscription level as well as the proliferation while they promot theapoptosis level of SGC7901cells. Compared with that of transmittingASODNs separately,the effect manifests itself more significantly with thecombination of NRP-1ASODN and VEGFR-2ASODN.
