PART ONEOBJECTIVE: To observe the effect of bcl-2 and c-erbB-2 genes antisense oligodeoxynucleotides (ASODN) combined transfection to the proliferation of the human nasopharynfeal carcinoma (NPC) HNE-1 cell lines. METHODS: There were 7 groups in our study, normal control groupâ… , c-erbB-2 sense mixed lipofectin (Lip) group â…¡, bcl-2 sense mixed Lip group â…¢, c-erbB-2 antisense mixed Lip groupâ…£, bcl-2 antisense mixed Lip group â…¤, whole-dose bcl-2 and c-erbB-2 antisense mixed Lip group â…¥, half-dose bcl-2 and c-erbB-2 antisense mixed Lip group â…¦. Cell cycle and protein expressing level were detected by flow cytometry and changes of the cell morpholog and the cell proliferation were observed by electronic microscope, MTT assay and cloning test. RESULTS: The trend of elongating S phase was the most obvious and the change of the cell morpholog was typical in half-dose combining group. The evident difference had been found about the inhibitory rate of cell proliferation between the antisense experimental group and other groups especially in half-dose combining group. The protein expressing level was decreased remarkably in the antisense experimental group especially in half-dose combining group. CONCLUSIONS: bcl-2 and c-erbB-2 genes ASODN combined transfection can inhibite the proliferation of the human nasopharynfeal carcinoma (NPC) HNE-1 cell lines and decrease the protein expressing level of bcl-2 and c-erbB-2.
