Objective:The basic studies showed that Pigment epithelium-derived factor (PEDF) plays an important role in inflammation. This study aims to investigate the association of serum PEDF with high-sensitivity C-reactive protein (hs-CRP) in women with polycystic ovary syndrome (PCOS).Methods:Ninety-six PCOS patients and63healthy controls (age-matched) were enrolled. Serum levels of PEDF, hs-CRP, Complement C3(C3), glucose, insulin, lipids and total testosterone were determined. hyperinsulinemic-euglycemic clamp is used to assess insulin sensitivity in96PCOS women and20controls.Results:Compared to the controls, serum PEDF and hs-CRP in PCOS patients were significantly higher. In PCOS women, PEDF positively correlated with body mass index (BMI), hs-CRP, C3, low-density lipoprotein cholesterol (LDL-c), and systolic blood pressure (SBP), and negatively correlated with M value and high-density lipoprotein cholesterol (HDL-c). After controlling for BMI, PEDF was still positively correlated with hs-CRP. Further more, regression analysis showed that hs-CRP, SBP, LDL-c and M value were independent determinants of serum PEDF in PCOS patients.Conclusions:The relationship between serum PEDF and hs-CRP are close in PCOS patients. Objective:To observe the effect of Tumor Necrosis Factor-α (TNF-α) intervention on pigment epithelium-derived factor (PEDF) expression and secretion in3T3-L1adipocytes.Methods:3T3-L1preadipocytes were cultured and diferentiated into the mature adipocyte.3T3-L1adipocytes were incubated with TNF-α at different concentrations (0.1,1,10and100ng/ml) for different periods of time (3,6,12and24h). PEDF mRNA expression was detected by Real-time Quantitative PCR. Total protein of PEDF secreted by these adipocytes was evaluated by ELIS A.Results:(1) Treatment of3T3-L1adipocytes with TNF-α at diferent concentrations decreased PEDF mRNA expression and secretion along with the elevated TNF-α concentration in the culture medium (P<0.05).(2) The inhibitive effect of TNF-α(10ng/ml) on PEDF mRNA expression gradually strengthened with the increase of treat time, and reach the strongest at12h (P<0.05).(3) Treated with TNF-α(10ng/ml), the inhibition rate of their secretion of PEDF was15%in the first6h,33%in the first24h (P<0.05).Conclusion:TNF-α significantly decreased the expression and secretion of PEDF in3T3-L1adipocytes in a dose-and time-dependent manner.
