The Anti-tumor Activity Of HepG2Tumor Vaccine Transfected GM-CSF Gene

Objective: In this study,we investigate the anti-tumor effects and mechanism ofHepG2tumor vaccine transfected GM-CSF gene，to accumulate data for clinicalapplication of tumor vaccine.Methods: Health human peripheral blood lymphocytes were isolated from peripheralblood of healthy donors and intraperitoneal injected into SCID mice at2x10⁷/ml.Simultaneously, hepatocellular carcinoma cells were intracutaneously injected intoSCID mouse at2x10⁶/ml. Ten days later,when the xenografted to about a（95.72±10.96） mm³, mice was randomly divided into3groups: Group Ⅰ,HepG2cells transfected with GM-CSF Gene after60Co irradiation group; Group Ⅱ, HepG2cells after⁶⁰Co irradiation group; Group Ⅲ, normal saline group. Every group had8mice. Every seven days,each mouse in GroupⅠ、Ⅱ、Ⅲ was respectively injected with0.2m（l1x10⁷/ml）HepG2cells transfected with GM-CSF Gene after⁶⁰Co irradiation、0.2ml（1x10⁷/ml）HepG2cells after60Co irradiation、0.2ml Physiological saline inthe medial skin of hind.Repeat three times.Observe the volume of subcutaneoustransplantation tumor in each growp. Seven days after the third injection,kill all themice,take the spleen and the blood.Cytotoxic rates were detected by using MTT assay.The level of IL-12and IL-4in serum was detected by ELISA method.Results: The volume of subcutaneous transplantation tumor in group HepG2tumorvaccine transfected hGM-CSF gene were smaller than HepG2cells after⁶⁰Coirradiation group and normal saline group,The activaty of CTL were significantlyhigher and the difference was significant （P<0.05）, Serum level of Th1-type cytokine IL-12increased after vaccination, and Th2-type cytokine IL-4s reducedafter vaccination，the differences were significant （P <0.05）.Conclusion: The HepG2tumor vaccine transfected GM-CSF gene can induce immuneresponse，have a certain anti-tumor activity.
