Objective:The rabbit stenosis model of vascular anastomosis were established at carotidartery, and local transfected caveolin-1by plasmid,Through observe the proliferationof vascular anastomosis.To study the role of caveolin-1in vascular anastomoticstenosis and the relationship to ERK1/2signaling pathway.Method:40adult rabbits were obtained from the Animal Center of the Nanhua University,builded the stonsis model of vascular anastomosis at carotid artery,and transfectedcaveolin-1by plasmid.The rabbits was divided them into4groups: normal group、surgical group、 blank transfected group and transfected group. Carotid arteryspecimens were taken for westernblot and PCR at7days,detect the expression ofprotein and mRNA. The rest specimens were taken for HE and immunohistochemicalat28days.Result:1. HE staining of tissue slice showed that: compared with normal group,surgicalgroup was significantly proliferation of intima, vessels wall thickening,luminalnarrowing; According to the intimal/medial area rate, transfected groupwas significantly lower than the surgical group,demonstrated that caveolin-1inhibitsstenosis of vascular anastomosis.2. RT-PCR detected the caveolin-1and ERK1genetranscription, the results show that: compared with the normal group, surgical groupand the blank transfected group caveolin-1mRNA was significantly lower(P<0.01);show that the expression of caveolin-1mRNA was lower.While sugicalgroup ERK1mRNA significantly higher,the exptession in transfected group waslower.3. Immunohistochemical detected the expression of caveolin-1, the resultsshow that: compared with normal group, the surgical group and blank transfected group caveolin-1expression was significantly lower. WesternBlot detected theexpression of caveolin-1protein,compared with normal group, the surgical groupcaveolin-1protein expression decreased significantly (P <0.05), indicating thatvascular injury can down-regulate the expression of caveolin-1;while transfectedgroup caveolin-1expression was significantly increased, indicating the success ofcaveolin-1transfection.Immunohistochemistry detect the phosphorylation of ERK1/2expression: compared with normal group, the surgical group and the blanktansfected group,the expression of phosphorylation of ERK1/2was significantlyhigher;while transfected group was significantly lower;western blotanalysis further confirmed the same trend. Show that caveolin-1can inhibit vascularanastomotic stenosis may be related to inhibition of ERK1/2activation.Conclusion1、 Caveolin-1can inhibit the rabbit neointimal proliferation after carotid arteryanastomotic.2、Caveolin-1inhibit anastomotic stenosis may be through regulate the activation ofERK...
