Objective: To explor of the role of JNK-paxillin signaling pathwaysystem inthe apoptosis of human colon cancer cell line (HT-29) induced by caffeic acidphenethyl ester (CAPE).Methods: The human colon cancer cell line HT-29was treated with differentcon-centrations of CAPE.MTT assay was used to observe the growth inhibition ratioof different con-centrations of CAPE effected on HT-29cells at differenttimes.Western blot was used to detect the expression of JNK and paxillin protein ofHT-29cells treated with different con-centrations of CAPE.Results: CAPE inhibited the proliferation of HT-29cell in a concentration andtime-dependent manner. MTT assay showed that the inhibited growth rate60.03%ofHT-29cells treated with10μg/mL CAPE for about72h was higher than the group28.93%which treated with2.5μg/mL CAPE for about72h (P<0.01),or the group36.74%which treated with10μg/mL CAPE for about24h(P<0.01).Western blottingexhibited that the expression of protein JNK/paxillin decreased following theconcentration of CAPE increasing.Treated with10μg/mL CAPE,the IOD of JNK was230.95±5.98and the IOD of paxillin was446.30±50.They both were lower than thegroup which Treated with2.5μg/mL CAPE.(P<0.01)。Conclusion: CAPE can inhibit the proliferation and induce the apoptosis ofhuman colon cancer cell line HT-29in vitro.The mechanism may be related withdown-regulated JNK/paxillin。...
