Effect Of Hsa-miR-100on Proliferation, Invasion And Apoptosis Of T24Bladder Cancer Cells | | Posted on:2013-01-03 | Degree:Master | Type:Thesis | | Country:China | Candidate:Y F Zhang | Full Text:PDF | | GTID:2234330374479518 | Subject:Surgery | | Abstract/Summary: | PDF Full Text Request | | Objectives:Upregulated hsa-miR-100in bladder cancer T24cells,we explored theinfluence ability of proliferation, invasion and apoptosis.Methods: Application chemical method synthetized hsa-miR-100mimics.Transiently transfected into T24cells and then used quantitative polymerase chainreaction(Q-PCR) detecting the relative expression of hsa-miR-100in T24cells.Makesure that the T24cells had been successfully transfection and on a high rate.Proliferation, invasion and apoptosis of the T24cells were respectively evaluatedusing MTT experiment, transwell assay and flow cytometer(FCM) test.Results:1﹑The bladder cancer T24cells could be transfected by hsa-miR-100mimics and then used quantitative polymerase chain reaction(Q-PCR) detecting therelative content was higher than the others (P<0.05).2﹑Compared transfectedhsa-miR-100mimics group with the control group and the blank group that thehsa-miR-100mimics group could significantly inhibit cell proliferation.(P<0.05).3﹑In the transwell assay we compared the experimental group(304.75±67.45)with thecontrol group(504.75±52.64)and the blank group(458.75±49.87)that thehsa-miR-100mimics group could significantly inhibit cell invasion of the bladdercancer T24cells(P<0.05).4﹑In the FCM test we put each group into the flowcytometric and found that the apoptosis rates of the experimental group, the controlgroup and the blank group respectively was(12.57±0.39)%,(7.71±0.31)%and(7.82±0.13)%. The apoptosis of T24cells obviously increased (P<0.05).Conclusion:Mature mimics of hsa-miR-100could transiently transfected intobladder cancer T24cells. The expression rise of hsa-miR-100significantlysuppressed proliferation force and invasion power of T24cells, and can accelerate T24cells apoptosis.
