| ObjectiveTo investigate whether5,7-dimethoxyflavone (5,7-DMF), a novel synthetic chrysin(5,7-dihydroxyflavone, chrysin, ChR) analogue, induces apoptosis in human lungcancer A549cell line, and explore whether the mechanism of this action is involved indepletion of intracellular glutathione(GSH).MethodsHuman lung cancer A549cell line was cultured in vitro. The apoptotic rate (thepercentage of sub-G1cells) was determined by flow cytometry (FCM) usingpropidium iodide (PI) fluorescence staining. DNA fragmentation was examined usingDNA agarose gel electrophoresis. The intracellular level of GSH was measured usingspectrophotometer.ResultsFCM with PI staining showed that5,7-DMF significantly increased the apoptoticrate of human lung cancer A549cells (P<0.05), in a concentration-dependentmanner. The ladder band could be observed in DNA agarose gel electrophoresis aftertreatment with5,7-DMF (30.0μmol/L) and0.3μmol/L taxol (Tax) for24h.5,7-DMFsignificantly reduced the intracellular level of GSH (P<0.05) in human lung cancerA549cells(P<0.05), in a concentration-dependent manner. Importantly, thepretreated with500μmol/L GSH for1h can effectively blocked the effects of5,7-DMF on induction of GSH depletion and apoptosis in lung cancer A549cells.Conclusion 1.5,7-DMF possesses the effects of induction of apoptosis in human lung cancerA549cells, in a concentration-dependent manner.2. Induction of apoptosis by5,7-DMF in human lung cancer A549cells was involvedin depletion of intracellular GSH. |
