The adverse reactions of Gegensu and Qingkailing injection are increasing recently with the wide use of them, of which the most serious may be allergic shock and deaths. A part of the allergy caused by the two injections occurred when given for the first time, and within30min after administration, which have the obvious characterisitics of anaphylactoid reactions.The clinical symptoms of anaphylactoid reactions are similar to those of type I allergic reactions, for example, nausea, vomiting, abdominal pain and diarrhea, urticaria, angioedema, allergic asthma and so on. However, its mechanisms are different from type I allergic reactions, so it is called "anaphylactoid reactions". Compared to type I allergic reactions, anaphylactoid reactions occur when the body contacts antigens for the first time without the accompany of immunoglobulin.Establishing evaluation models of anaphylactoid reactions to assess Qingkailing injection and Gegensu injection has great guiding significance for their use in the clinical. The evaluation models our experiments have established conclude allergic reaction detection model of anesthesia Beagle dogs and so on. This study considers improving the evaluation methods on the basis and build the evaluation models of anaphylactoid reactions of conscious Beagle dogs using the DSI telemetry system to assess the anaphylactoid reactions caused by Qingkailing injection or Gegensu injection; Isolating of canine peripheral blood mononuclear cells to evaluate the effect on cytokines caused by Qingkailing injection or Gegensu injection.1Establishing evaluation model of anaphylactoid reactions on conscious Beagle dogsObjective:To establish the evaluation model of anaphylactoid reactions on conscious Beagle dogs. Method:Imbedding the sensor in abdomen of dogs. Give Tween-80and Compound48/80to assess the practicability of the evaluation models of anaphylactoid reactions. Monitor the change in dogs with DataquestA.R.T.4.3system and study the sensitivity of blood pressure and heart rate as the indicators of anaphylactoid reactions. Result:After the administration of Compound48/80and Tween-80, blood pressure of dogs decreased and cardiac acceleration appears, and the anaphylactoid symptoms of disturbances, skin flushing, scleral hyperemia appear in the dogs. Conclusion:The evaluation models of anaphylactoid reactions on conscious Beagle dogs is successful and can be used to detect drug anaphylactoid reactions.2Assessment of the anaphylactoid reactions in conscious dogs caused by Qingkailing injection and Gegensu injectionObjective:To evaluate the anaphylactoid reactions caused by Qingkailing and Gegensu using the evaluation model we have built. Method:Give the conscious dogs Qingkailing and Gegensu injection of different dosages intravenously under120mL-h"1, and monitor the change in dogs with DataquestA.R.T.4.3system. Result: Changes of blood pressure or heart rate in dogs were not apparent after the administration of Qingkailing injection of1.2mL·kg-1,2.3mL·kg-1,11.5mL·kg-1or Gegensu injection of0.24mL·kg-1. Blood pressure in dogs decreased after the administration of Gegensu injection of0.47mL·kg-1. Average blood pressure dropped from (115.35±7.73) mmHg to (91.74±10.18) mmHg (n=4,P<0.01) and systolic blood pressure dropped from (147.33±14.65) mmHg to (112.88±6.51) mmHg(n=4,P<0.01) thirty minutes later, but obvious signs of change of the dogs are not observed. Conclusion:Qingkailing injection don’t cause anaphylactoid reactions in conscious Beagle dogs under120mL·h-1. Gegensu injection can decrease the blood pressure in conscious Beagle dogs, which is related to the dosage.3Evaluation of the effect on the cytokines of dogs caused by Qingkailing injection and Gegensu injectionObjective:To study the effect on the cytokines of dogs caused by Qingkailing injection and Gegensu injection and assess the relationship between anaphylactoid reactions and cytokines. Method:PBMC are separated by density gradient centrifugation, then react with Tween80, Compound48/80, Qingkailing injection and Gegensu injection. The cytokines are assayed using corresponding ELISA kits. Result: Compared to the control group, the concentration of TNF-α and IFN-γ released by PBMC decreased after the reaction with all the drugs, all of the OD results of IL-6are lower than the minimum value of the standard value. The results are not consistent with that of the literatures. Conclusion:Tween80, Compound48/80, Qingkailing injection and Gegensu injection can decrease the release of TNF-α and IFN-γ.
