Objective:In the article, on the based of supramolecular theory, the Houttuynia injection and ShuangHuangLian injection, as the model medicals, were studied the immunotoxicity effects owning to adjuvant material, adjuvants and method of administration, combined with the behavioral science investigation, biochemical indicator detection and immune fingerprint method, in order to explore the correlation between supramolecular and anaphylactoid reaction, and to improve techniques of screening anaphylactogen directionly. This study would benefit to illustration of sensitized mechanism for Chinese Medicine (CM) injections.Methods:The evaluation of animal behaviors and leakage amount of auricle Evans blue, combined with the detection of HIS、CH50、TPS、IgE were utilized to estimate the influent differences on anaphylactoid reaction caused by the tween-80 as adjuvant material formed Houttuynia injection, the Freund’s adjuvant combined with two injections, and the method of administration for two injections as well. The immune fingerprint method, was adopted for determination of ingredients in Houttuynia injection with GC/MS and ShuangHuangLian injection withHPLC/MS. According to their immune fingerprint differences, the mechanisms of anaphylactoid reaction have been illustrated. The experiment were taken Kunming mice and Wistar rats as sensitized animals, and introduced supramolecules theory to investigation their correlation to anaphylactoid reaction.Results:(1)The aizen ratios on administration of injection of Compound48/80, Houttuynia injection, Houttuynia distillate, tween-80,saline into mice, were 100.00%,100.00%,83.30%,100.00%, 0.00% and on administration of intragastric 83.30%,50.00%,75.00%, 83.30%,0.00% respectively, in which the differences were statistically significant compared with saline control group(P<0.01, P<0.05). (2) Score of different method of administration about aizen rate on mice: injection group> filtration injection group> lavage group, difference between groups was statistically significant (P<0.05). When self-made syringe injected (i.e., filtration injected) were ultilized among Compound48/80, Houttuynia injection, ShuangHuangLian injection compared with ordinary disposable syringe injected, the increase rate of leakage quantity of Evans blue on mice’s auricle decreased to 90.8%, 72.7%,83.8%. (3) Compared with group added freund’s adjuvant, blood index such as HIS, TPS, IgE levels of single injection group and individual adjuvants group, were significantly higher (P<0.01), but increased levels of CH50 is not subtle. Contrasting YXC+Z group with YXC, according to different level, such as HIS, CH50, TPS, IgE, multiplesihs of adjuvant enhanced were respectively 1.86,1.36,1.24, 2.78 times. SHL+Z group compared to the SHL group,respectively 1.64, 1.91,1.30,3.13 times. (4) Common syringe dosing, self-made syringe dosing and lavage for medicine, compound, YXC, SHL group compared with saline group, the experimental data differences between groups were statistically significant (P< 0.01, P< 0.01, P< 0.05). The filtration injected compared with injected:the level of filtration reducing injection ratio of HIS,CH50, TPS and IgE is 0.92,0.64,0.91; 0.89,0.59,0.84; 0.90,0.75, 0.86; 0.96,0.82,0.90.(5) There are 22 kinds of composition in ShuangHuangLian injection and its medicated plasma can be absorbed by specific IgE antibody, their molecular weight range from 114 to 908, they are mostly acids, esters and nitrogen compounds. There are 13 kinds of composition in Heartleaf houttuynia herb injection and its medicated plasma can be absorbed by specific IgE antibody, their molecular weight range from 128 to 340, they are mostly fat ketones, esters, alcohols, etc.Conclusion:(1) Anaphylactoid caused by Houttuynia injection is related to adjuvant material Tween-80, and different administrations can affect the sensibilization. Sensibilization:injections> filtration injections >intragastric administration. In this study, using self-made injector can reduce sensitization of TCM injection.(2) In evaluation of anaphylactoid, animals vascular permeability increase, which use Evans blue as the tracer, through measuring the effusion of tissues Evans blue to evaluate the strength of the allergic reactions to animals, the result of Auricle aizen experiment is easily influenced by subjective factors, Suggestting combining with HIS, CH50, TPS, IgE blood index to test. (3) For Houttuynia injection, the enhancement effect of adjuvants for HIS and IgE is stronger, and for ShuangHuangLian injection,the enhancement effect of adjuvants of CH50 and IgE is stronger. Self-made injector can reduce the levels of rats HIS, CH50, TPS, IgE (P<0.05), which is compound48/80 group, YXC group and SHL group, and the reduction effect for YXC group is stronger than the SHL group (P<0.01), which show the special sintered layer at the top of the cylinder structure can effectively dismantled supramolecular sensitization. (4) By immune adsorption experiments, combining with IgE antibody adsorption anterior-posterior of ShuangHuangLian injection of HPLC fingerprint and Houttuynia injection of GC fingerprint, can be further confirmed that the different administrations can change sensitization of TCM injections, through specific IgE antibody adsorption resistance could capture supramolecular anaphylactogen successfully. (5) The acids, esters and nitrogen compounds in ShuangHuangLian injection and and its medicated plasma, the fat ketone, esters, alcohols in heartleaf houttuynia herb injection and its medicated plasma.can form supramolecular hapten, so as to make TCM to cause allergic reactions when it is used as injections, but not as oral preparations.
