Objective To detect the protein and mRNA expression of TBX2、 PAX9in tissues fromendometrioid adenocarcinoma, endometrial hyperplasia and normal endometrium. To investigatetheir roles in carcinogenesis, biological behavior and evaluate their clinicopathologic significance.Methods The expression of TBX2, PAX9proteins in normal tissues (n=30), endometrialhyperplasia tissues (n=60) and endometrial adenocarcinoma tissues (n=82) were examined by tissuemicroarray technique(TMA), immunohistochemistry (SP method) and in situ hybridization method.Results1.The positive rates of TBX2,PAX9protein and mRNA in endometriod adenocarcinomawere respectively76.8%(63/82),73.2%(60/82) and72.0%(59/82),69.5%(57/82).2.The positiverates of TBX2protein and mRNA in normal endometrium, endometrial simple hyperplasia,endometrial atypical hyperplasia and endometrial carcinomas were gradually increased (p<0.01).The positive rate of TBX2were related to the histological grade (p<0.01), FIGO stage (p<0.05),depth of invasion (p<0.01), not to the lymph node metastasis (p>0.05).3.The positive rates ofPAX9protein and mRNA in endometrial carcinomas were higher than normal endometrium,endometrial simple hyperplasia (p<0.01), but not related to the positive rates in endometrialatypical hyperplasia (p>0.05). The positive of PAX9were related to the histological grade (p<0.01),FIGO stage (p<0.01), depth of invasion (p<0.05), not to the lymph node metastasis (p>0.05).4.The expression of TBX2、PAX9protein were positively related their mRNA and the expression ofTBX2were positively related to that of PAX9(rs=0.427, p<0.01). Conclusion1.The expressionof TBX2in endometrioid adenocarcinoma was higher than that in normal endometrium andendometrial hyperplasia,which promoted cell proliferation and associated with the development andprogression of endometrioid adenocarcinoma.2. The expression of PAX9were upregulated in complex endometrial hyperplasia with atypical hyperplasia as well as endometrioid adenocarcinomatissues, implying that PAX9were activated in the early stage of endometrioid adenocarcinoma.3.It reveal that the abnormal expression of TBX2, PAX9played a role in the development andprogression of endometrial adenocarcinoma, and TBX2may associate with regulations of PAX9.4.The results of this study support the assume that PAX9, TBX2may be important biomarkers in thedetection of biological behavior and prognosis in endometrial adenocarcinoma.
