Objective Tumstatin is composed of three helical regions in the middle of the α3chainof type IV collagen C-terminal12amino acids and non-collagenous1(NC1) of232amino acids,244amino acids. Neovascularization and tumor xenograft model studieshave shown that recombinant tumstatin significantly inhibit the proliferation ofendothelial cells specifically induces apoptosis of endothelial cells, thereby inhibitingtumor growth in vitro and in vivo. Tumstatin is endogenous neovascularization inhibitor,in vivo changes in tumor growth and metastasis. In this study, with the Taqmantechnology of fluorescent quantitative reverse transcription polymerase chain reaction(FQ-RT-PCR), tumstatin/type IV collagen α3chain [α3(IV)] mRNA expression levelsof the quantitative were detected in lung cancer. Corresponding tissue tumstatin/typeIV collagen α3chain protein expression levels with immunohistochemistry andWestern-Blot were detected. Tumstatin/α3(IV) of changes in gene expression andprotein expression between the two clinical value in the development of lung cancerwere evaluated in patients with lung cancer..Methods①48cases of confirmed initial surgical resection of lung cancer patients forlung cancer tissue and adjacent normal tissue were collected.②In the α3(IV) NC1domain coding sequence a pair of primers and a Taqman probe; optimize the conditionsof the reaction system standards and threshold cycle (Ct value) of different plasmidDNA standard curve, detection in lung cancer α3in (IV) mRNA levels; andmethodological evaluation.③Lung cancer and paraneoplastic tissue in tumstatin/α3(IV) protein levels were detected by protein immunoblot (Western-Blot) and immunohistochemical, explore the lung cancer the relationship between thedevelopment of its protein expression.④CD31monoclonal antibody detected lungcancer tissues and adjacent tissues of the vascular density MVD analysis tumstatin/α3(IV) and MVD.Results α3(IV) mRNA expression was detected in real-time fluorescent quantitativePCR method, the detection sensitivity of800copies/μl; a good linear relationship withthe Ct value between103~108copies/μl; PCR amplification efficiency99.4%; α3(IV)mRNA expression quantitatively was detected in48cases of lung cancer patients andα3(IV) mRNA of lung cancer group were significantly lower than the cancer adjacenttissues, the difference was statistically significant (P<0.01) in lung cancer tumstatinprotein expression down/deletion rate was significantly higher than the cancer adjacenttissues (P<0.01). Clinical monitoring of lung carcinoma treated with α3(IV) mRNAcontent detection and TNM staging of the degree of correlation (P<0.05).Conclusions Microvessel density are closely related in the expression of in conclusiontumstatin/α3(IV) and lung cancer, lung cancer tissue tumstatin/α3(IV) of the expressionof genes with low expression is one of the protein missing an important mechanism thatmay occur in lung cancer development process playing an important role, which can beused as effective molecular markers.
