ObjectiveTo investigate the clinical application of platelets, serum and platelet-poor plasma(PPP) vascular endothelial growth factor(VEGF), Tumstatin and TSP-1determination innon-small cell lung cancer (NSCLC).MethodsAnticoagulant whole-blood samples of24NSCLC patients and24sex-andage-matched healthy controls were collected by venipuncture into CTAD (citrate,theophylline, adenosine and dipyridamole) tubes for platelet and PPP recovery, andserum samples were retrieved by blood collection without the addition of anticoagulants.Peripheral blood samples from NSCLC patients were drawn within1week beforesurgery or chemotherapy. The control subjects of the study had no history of malignancy,inflammatory disease, diabetes mellitus, or non-steroidal anti-inflammatory drug use.The tumor tissue pathological slices were derived from the Pathology Department aftersurgery.â‘ The concentrations of proteins of interest in platelet, serum and PPP weremeasured using an Enzyme-Linked Immunosorbent Assay (ELISA).â‘¡Microvesseldensity (MVD) expression in tumor tissue from24cases of lung cancer were examinedby immunohistochemistry methods.ResultsPlatelet and serum VEGF levels were higher in NSCLC patients compared tocontrols (P<0.01and P<0.05, respectively). Conversely, NSCLC patients exhibitedlower platelet/serum tumstatin and TSP-1levels than controls (P<0.05, respectively).However, PPP VEGF and TSP-1levels in all participants were below the detection limit of the assay. And there was no significant difference in the PPP tumstatin concentrationsbetween patients and controls (P>0.05). Receiver-operating characteristics (ROC) curveanalysis for VEGF, tumstatin and TSP-1in serum had an area under the curve (AUCranging from0.772to0.832, P<0.01), but platelet angiogenic factors were superior(AUC ranging from0.831to0.969, P<0.01). The levels of platelets and serum VEGFand MVD count in cancer tissue were increased, the levels of platelets/serum Tumstatinand TSP-1were decreased in lung cancer patients with stage; There was a statisticallysignificant positive correlation between platelet/serum VEGF levels and MVD (P<0.01and P<0.05, respectively). However, negative correlation were found betweenplatelet/serum tumstatin and TSP-1levels and MVD (P<0.05), respectively.ConclusionThe platelet-derived angiogenesis regulators can be used as relevant biomarker toreflect human tumor angiogenesis and progression compared with the angiogenesisregulators in serum. The balance between Tumstatin/TSP-1and VEGF in peripheralblood of lung cancer play an important role in the regulation of angiogenesis. The levelsof angiogenesis regulators in peripheral blood is closely correlated with MVD, whichcould play an critical role in monitoring the condition of NSCLC.
