| ObjectiveHepatocyte growth factor has angiogenic,anti-fibrosis,anti-inflamminationbiological functions. The aim of our study was to investigate the therapeutic effect onattenuating left vetricular remodeling (LVR) after given adenovirus-mediated humanhepatocyte growth factor (Ad-HGF) transmyocardial injection. Test LVMIã€CVFã€LVEDPã€Â±dp/dtmax. Detect the vessal density of myocardial;HGFand NF-κB P50expressions in myocardium; ELLSA analysis of serum IL-1, CRP,TNF-α.MethodsTotally twenty-four SHR were randomly divided into three groups:(1)(SHR-NS group, normal saline,0.1ml);(2)(SHR-Ad group, null-,Ad5,0.1ml);(3)(SHR-AdHGF group1x109Pfu/ml Ad5-HGF,0.1ml)ï¼›(4)(Control group) consist of8Wista-Kyoto (WKY) rats. Salineã€Null-Ad or Ad-HGF was transfected into the leftventricle wall at five different points by direct injection respectively,a left anterolateralthoracotomy was performed to visualize the LV free wall.4weeks after surgery,testLVMIã€CVFã€LVEDPã€Â±dp/dtmax.Immunohistochemical examinationfor CD34;Western blot analysis of HGFand NF-κB P50of myocardium; ELLSA analysis of serumIL-1, CRP,TNF-α.Results (1) Compared with the Control group,LVMIand CVF significantly increasedin SHR-NS group (P<0.05);Compared with the SHR-NS group,LVMI and CVFshowed a significant decrease in the Ad-HGF group (p<0.05).(2) Compared with theSHR-NS group,LVEDP showed a significant decrease in the SHR-AdHGF group (p<0.05);. Compared with the SHR-NS group,±dp/dtmax significantly increased inAd-HGF group group (3) Compared with the SHR-NS group(5.13±1.09),the vessaldensity of myocardial significantly increased in theSHR-AdHGF group(10.78±1.29)(p<0.05).(4) Compared with the Control group, the serum of IL-1, CRP,TNF-α andmyocardial NF-κB proteins expressions significantly increased in theSHR-NS group(p<0.05);Compared with the SHR-NS group,the serum of IL-1, CRP,TNF-α andmyocardial NF-κB proteins expressions significantly reduced in theSHR-AdHGF group(p<0.05).Conclusions Our results demonstrated that gene transfection of Ad-HGF attenuatedthe left vetricular remodeling. Our study has proved that Ad-HGF hasangiogenic,anti-inflammination biological functions.That is why Ad-HGF couldattenuate the left vetricular remodelling. |
