Objective:In order to provide some basic exploration for the development and application of the total flavonoids of wild kudzu and the innovation of Chinese medicine, we conducted systematic pharmaceutical experiments and studied on the medicinal value of wild kudzu and total flavonoids of wild kudzu extract kudzu.Methods:We conducted chemical separation of wild kudzu by the separation and purification methods of normal phase and reversed-phase silica gel column chromatography, gel filtration, recrystallization, and preparative TLC, we conducted content determination of total flavonoids and daidzein of wild kudzu by UV and HPLC; Determined the optimum conditions of wild kudzu by orthogonal test, wild kudzu extract as these samples,establishing HPLC fingerprints and quality standards of wild kudzu extract; we conducted the single factor of accessories and tablet on wild kudzu extract, in order to determine the wild kudzu dispersible tablets of the preparation process.Results:Field resource investigation results showed that, wild kudzu resources are widely distributed within the scope of Shandong Province, The active ingredient content in wild kudzu approximation of kudzu root and the vines contained a higher content of daidzein, the active ingredient content of wild kudzu in jinan region is higher than other regions; A variety of separation and purification means to isolate the chemical constituents contained in,and structural analysis of each component physical and chemical properties and its NMR data,13compounds isolated from the wild kudzu extract; Orthogonal design wild kudzu optimum extraction process for6times amount of70%ethanol to operate heating circumfluence, for3times and1.5h per time, total flavonoid content of up to23.11%, daidzein was5.62%; We conducted study of fingerprints on wild kudzu extract by HPLC, isolated17total bee, whether it is the relative retention time or relative peak area differences were small; Inspect disintegrant, glidant, filler, lubricant in dispersible tablet of wild kudzu extract, preferably a cross-linked povidone, silica powder, microcrystalline cellulose, magnesium stearate as its accessories, φ12mm light concave stamping film, adjust tablet weight of about500mg,hardness control in5~6kg, dispersible tablets disintegrating in the110s.Conclusion:By the pharmaceutical research on wild kudzu,the results showed that wild kudzu has great medicinal value for development, the study can provide the basis of the basic research for exploring the development of wild kudzu and resource utilization; Through the preparation process of kudzu dispersible tablets, provide an effective basis for wild kudzu extract research and development into innovative new drugs.
