Observation Of The Effect Of Fluorofenidone On Rat Hepatic Stellate Cell Proliferation

ObjectiveTo investigate the effect of fluorofenidone on the proliferation of rat hepatic stellate cell and its mechanism.Method1. primary stellate cells were isolated from livers of S-D rats by perfusion with pronase and collagenase.2. After isolated and cultured for14days, cells were divided into four groups:normal group、model group(10ng/ml PDGF stimulation)>200μg/ml AKF-PD treatment group and400μg/ml AKF-PD treatment group.Cells were pretreated with drugs for24hours before stimulation with PDGF(10ng/ml) for24hours.MTT assay was used to observe the effect of AKF-PD on HSC proliferation.3. After isolated and cultured for14days, cells were divided into four groups:normal group、model group(10ng/ml PDGF stimulation)、400μg/ml AKF-PD treatment group and400μg/ml PFD treatment group. Cells were pretreated with drugs for24hours before stimulation with PDGF(1Ong/ml) for24hours.Proteins were extracted and studied by Western Blot for CyclinD1、CyclinE、P27Kip1expression.Results1. Compared with normal group,the rate of growth of hepatic stellate cell was higher in the model group;compared with model group,the growth rate of hepatic stellate cell were significantly reduced in the AKF-PD treatment groups with different concentrations.2. Compared with normal group,the expression level of CyclinDl and CyclinE were higher,and that of P27Kip1was lower in the model group;compared with model group, the expression level of CyclinDl and CyclinE were lower,and that of P27Kip1was higher in the AKF-PD treatment group.Conclusion1. AKF-PD inhibited the proliferation of primary hepatic stellate cell significantly.2. The inhibitory effect of AKF-PD on primary hepatic stellate cell proliferation was possibly due to downregulation of CyclinD1、CyclinE expression and upregulation of P27Kip1expression.
